1Isolation and Identification of Cancer Stem-Like Cells from Murine Melanoma Cell Lines显示文摘In current study,cancer stem-like cells in the murine melanoma B16F10 cells were investigated.CD phenotypes of the B16F10 cells were analyzed by flow cytometry,and the specific CD phenotype cells from the B16F10 cells were isolated by MACS.Then we used colony formation assay in soft agar media,the cell growth assay in serum-free culture media as well as the tumorigenicity investigation of the specific CD phenotype cells in C57BL/6 mice, respectively,to identify cancer stem-like cells in the B16F10 cells.The results showed that the B16F10 cells could form spherical clones in serum-free culture media,and the rate of clonegenesis of CD133 + ,CD44 + and CD44 + CD133 + cells was higher than that of CD133 - ,CD44 - and CD44 + CD133 - cells in soft agar media,respectively. The tumorigenic potential of CD133 + ,CD44 + ,CD44 + CD133 + cells and CD44 + CD133 + CD24 + cells was stronger than that of CD133 - ,CD44 - ,CD44 + CD133 - cells and CD44 + CD133 + CD24 - cells in mice,respectively.In conclusion,the CD44 + CD133 + CD24 + cells have some biological properties of cancer stem-like cells or are highly similar to the characteristics of cancer stem cells(CSC).These results provide an important method for identifying cancer stem-like cells in B16F10 cells and for further cancer target therapy.Jun Dou Meng Pan Ping Wen Yating Li Quan Tang Lili Chu Fengshu Zhao Chuilian Jiang Weihua Hu Kai Hu Ning Gu 2007Cellular & Molecular Immunology2007,4,6:25

3Liver metastasis from hepatoid adenocarcinoma of the stomach mimicking hepatocellular carcinoma: Dynamic computed tomography findings显示文摘AIM: To evaluate the dynamic computed tomography(CT) findings of liver metastasis from hepatoid adenocarcinoma of the stomach(HAS) and compared them with hepatocellular carcinoma(HCC).METHODS: Between January 2000 and January 2015, 8 patients with pathologically proven HAS and liver metastases were enrolled. Basic tumor status was evaluated for the primary tumor location and metastatic sites. The CT findings of the liver metastases were analyzed for tumor number and size, presence of tumor necrosis, hemorrhage, venous tumor thrombosis, and dynamic enhancing pattern.RESULTS: The body and antrum were the most common site for primary HAS(n = 7), and observed metastatic sites included the liver(n = 8), lymph nodes(n = 7), peritoneum(n = 4), and lung(n = 2). Most of the liver metastases exhibited tumor necrosis regardless of tumor size. By contrast, tumor hemorrhage was observed only in liver lesions larger than 5 cm(n = 4). Three patterns of venous tumor thrombosis were identified: direct venous invasion by the primary HAS(n = 1), direct venous invasion by the liver metastases(n = 7), and isolated portal vein tumor thrombosis(n = 2). Dynamic CT revealed arterial hyperattenuation and late phase washout in all the liver metastases.CONCLUSION: On dynamic CT, liver metastasis from HAS shared many imaging similarities with HCC. 6Immunogenicity and safety of a severe acute respiratory syndrome coronavirus 2 inactivated vaccine in healthy adults: randomized, double-blind, and placebo-controlled phase 1 and phase 2 clinical trials显示文摘Background:The significant morbidity and mortality resulted from the infection of a severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)call for urgent development of effective and safe vaccines.We report the immunogenicity and safety of an inactivated SARS-CoV-2 vaccine,KCONVAC,in healthy adults.Methods:Phase 1 and phase 2 randomized,double-blind,and placebo-controlled trials of KCONVAC were conducted in healthy Chinese adults aged 18 to 59 years.The participants in the phase 1 trial were randomized to receive two doses,one each on Days 0 and 14,of either KCONVAC(5 or 10 mg/dose)or placebo.The participants in the phase 2 trial were randomized to receive either KCONVAC(at 5 or 10 mg/dose)or placebo on Days 0 and 14(0/14 regimen)or Days 0 and 28(0/28 regimen).In the phase 1 trial,the primary safety endpoint was the proportion of participants experiencing adverse reactions/events within 28 days following the administration of each dose.In the phase 2 trial,the primary immunogenicity endpoints were neutralization antibody seroconversion and titer and anti-receptor-binding domain immunoglobulin G seroconversion at 28 days after the second dose.Results:Inthe phase1 trial,60 participantswere enrolled andreceived at least one dose of 5-mgvaccine(n=24),10-mgvaccine(n=24),or placebo(n=12).In the phase 2 trial,500 participantswere enrolled and received at least one dose of 5-mg vaccine(n=100 for 0/14 or 0/28 regimens),10-mg vaccine(n=100 for each regimen),or placebo(n=50 for each regimen).In the phase 1 trial,13(54%),11(46%),and seven(7/12)participants reported at least one adverse event(AE)after receiving 5-,10-mg vaccine,or placebo,respectively.In the phase 2 trial,16(16%),19(19%),and nine(18%)0/14-regimen participants reported at least oneAEafter receiving 5-,10-mg vaccine,or placebo,respectively.Similar AE incidences were observed in the three 0/28-regimen treatment groups.No AEs with an intensity of grade 3+were reported,expect for one vaccine-unrelated seriousAE(foot fracture)reported in the phase 1 trial.KCONVACinduced significant antibody responses;0/28 regimen showed a higher immune responses than that did 0/14 regimen after receiving two vaccine doses.Conclusions:Both doses of KCONVAC are well tolerated and able to induce robust immune responses in healthy adults.These results support testing 5-mg vaccine in the 0/28 regimen in an upcoming phase 3 efficacy trial.Trial Registration:http://gffzz62a03ee25e6f4a77h0wukpb6vpxux6ufp.ffgz.tsg.suse.edu.cn/index.aspx(No.ChiCTR2000038804,http://gffzz62a03ee25e6f4a77h0wukpb6vpxux6ufp.ffgz.tsg.suse.edu.cn/showproj.aspx?proj=62350;No.ChiCTR2000039462,http://gffzz62a03ee25e6f4a77h0wukpb6vpxux6ufp.ffgz.tsg.suse.edu.cn/showproj.aspx?proj=63353).Hong-Xing Pan Jian-Kai Liu Bao-Ying Huang Gui-Fan Li Xian-Yun Chang Ya-Fei Liu Wen-Ling Wang Kai Chu Jia-Lei Hu Jing-Xin Li Dan-Dan Zhu Jing-Liang Wu Xiao-Yu Xu Li Zhang Meng Wang Wen-Jie Tan Wei-Jin Huang Feng-Cai Zhu 2021Chinese Medical Journal2021,,11:11
7Reversal of multi-drug resistance by pSUPER-shRNA-mdr1 in vivo and in vitro显示文摘AIM:To explore the possibility of reversing multi-drug resistance(MDR) to HepG2/mdr1 in vitro and in vivo with RNA interference(RNAi) . METHODS:HepG2/mdr1 was obtained by cloning the whole gene mdr1 into HepG2 cells.shRNA targeting sequence was designed to be homologous to the P-gp encoding MDR1 mRNA consensus sequence.pSUPERshRNA/mdr1 was constructed using the enzymedigested technique.HepG2/mdr1 cells were transfected with vectors of pSUPER-shRNA/mdr1 to measure their efficacy by real-time PCR for mdr1 mRNA,flow cytometry(FCM) for P-gp expression,and Rhodamine efflux,MTT method for HepG2/mdr1 function,respectively.In vivo,mice tumors were treated by injecting pSUPER-shRNA/mdr1 in situ and into intraabdominal cavity.Tumors were collected to create cell suspension and cryosections after chemothearpy with adiramycin and mytomycin.The cell suspension was incubated in RPMI-1640 supplemented with G418 to screen stable cells for appreciating the reversal of MDR. Cryosections were treated with immunohistochemistry technique to show the effectiveness of transfection and the expression of P-gp. RESULTS:pSUPER-shRNA/mdr1 was successfullyconstructed,which was confirmed by sequencing. The MDR phenotype of HepG2/mdr1 was decreased significantly in vitro transfection.HepG2/mdr1 showing its MDR was reversed notably in P-gp expression(11.0%vs 98.2%,P<0.01) .Real-time PCR showed that mRNA/mdr1 was lower in test groups than in control groups(18.73±1.33 vs 68.03±2.21,P<0.001) . Compared with HepG2,the sensitivity of HepG2/mdr1 and HepG2/mdr1-dsRNA cells to ADM was decreased by 1.64 times and 15.6 times,respectively. The accumulation of DNR in positive groups was decreased evidently.In vivo,the p-gp expression in positive groups was significantly lower than that in control groups(65.1%vs 94.1%,P<0.05) .The tumor suppressing rate in test groups was 57.8%. After chemotherapy,the growth rate in test groups was lower than that in control groups(700.14±35.61 vs 1659.70±152.54,P<0.05) .Similar results were also observed under fluorescence microscope,and confirmed by Image-Pro Plus 4.5 analysis. 8Overexpression of micro RNA_(40)8 enhances photosynthesis, growth, and seed yield in diverse plants显示文摘The ability of a plant to produce grain, fruit, or forage depends ultimately on photosynthesis. There have been few attempts, however, to study micro RNAs, which are a class of endogenous small RNAs post-transcriptionally programming gene expression, in relation to photosynthetic traits. We focused on miR_(40)8, one of the most conserved plant miRNAs, and overexpressed it in parallel in Arabidopsis, tobacco, and rice. The transgenic plants all exhibited increased copper content in the chloroplast, elevated abundance of plastocyanin,and an induction of photosynthetic genes. By means of gas exchange and optical spectroscopy analyses, we showed that higher expression of miR_(40)8 leads to enhanced photosynthesis through improving efficiency of irradiation utilization and the capacity for carbon dioxide fixation. Consequently, miR_(40)8 hyper-accumulating plants exhibited higher rate of vegetative growth. An enlargement of seed size was also observed in all three species overproducing miR_(40)8. Moreover, we conducted a 2-year-two-location field trial and observed miR_(40)8 overexpression in rice significantly increased yield, which was primarily attributed to an elevation in grain weight. 12Inhibitory effect of schisandrin B on free fatty acid-induced steatosis in L-02 cells显示文摘AIM:To investigate the effects of schisandrin B (Sch B) on free fatty acid (FFA)-induced steatosis in L-02 cells.METHODS:Cellular steatosis was induced by incubating L-02 cells with a FFA mixture (oleate and palmitate at the ratio of 2:1) for 24 h.Cytotoxicity and apoptosis were evaluated by 3-(4,5-dmethylthiazol-2-yl)-2,5-diphenyl tetrazolium bromide assay and Annexin V/propidium iodide staining,respectively.Cellular total lipid was determined using a photocolorimetric method after Nile red staining,and triglyceride content was measured using an enzymatic kit.To study the effects of Sch B on steatosis,L-02 cells were treated with Sch B (1-100 μmol/L) in the absence or presence of 1 mmol/L FFA for 24 h,and cellular total lipid and triglyceride levels were measured.To explore the mechanisms of action of Sch B in the steatotic L-02 cells,mRNA levels of several regulators of hepatic lipid metabolism including adipose differentiation related protein (ADRP),sterol regulatory element binding protein 1 (SREBP-1),peroxisome proliferator-activated receptor (PPAR)-α and PPAR-γ were measured by quantitative real-time polymerase chain reaction (PCR),and protein levels of ADRP and SREBP-1 were measured by immunoblotting.RESULTS:Treatment with 1 mmol/L FFA for 24 h induced intracellular lipid accumulation in L-02 cells comparable to that in human steatotic livers without causing apparent apoptosis and cytotoxicity.Sch B mitigated cellular total lipid and triglyceride accumulations in the steatotic L-02 cells in a dose-dependent manner.Quantitative real-time PCR and Western blot analyses revealed that treatment of L-02 cells with 100 μmol/L Sch B reverted the FFA-stimulated up-regulation of ADRP and SREBP-1.CONCLUSION:Sch B inhibits FFA-induced steatosis in L-02 cells by,at least in part,reversing the up-regulation of ADRP and SREBP-1.Jian-Hong Chu Hui wang yan ye Ping-Kei Chan Si-Yuan Pan Wang-Fun Fong Zhi-Ling Yu 2011World Journal of Gastroenterology2011,17,19:4
13Synergistic electronic interaction between ruthenium and nickel-iron hydroxide for enhanced oxygen evolution reaction显示文摘The efficiency of electrochemical water splitting is extremely hampered by the sluggish oxygen evolution reaction(OER)occurred at the anode.Therefore,developing high-performance OER electrocatalysts is crucial for realizing the industrialized application of water splitting.Herein,a high-efficiency electrocatalyst of ruthenium-decorated nickel-iron hydroxide(10 Ru-NiFe LDH)supported on Ni foam is successfully synthesized for OER.Modifying NiFe LDH with ruthenium can optimize the electronic density to form high valences of metal sites,which is beneficial to promote its OER performance.Consequently,the 10 Ru-NiFe LDH only needs a low overpotential of 222 mV to achieve a current density of50 mA,cm^(-2),which exhibits fast OER kinetics with a small Tafel slope of 58 mV.dec^(-1).Moreover,this electrocatalyst shows high stability over 20 h at a high current density of 100 mA·cm^(-2)without obvious decay.The decent OER performances can be ascribed to the increased active sites and the synergistic electronic interactions among Ni,Fe and Ru.This work provides an effective approach for designing desirable electrocatalysts for OER.Hao Cui Han-Xiao Liao Zhi-Lu Wang Jian-Ping Xie Peng-Fei Tan De-Wei Chu Pan Jun 2022Rare Metals2022,41,8:3

16Progress on major genes for high fecundity in ewes显示文摘The existence of major genes affecting fecundity in sheep flocks throughout the world has been demonstrated.Three major genes whose mutations can increase ovulation rate have been discovered,and all related to the transforming growth factorβ(TGF-β)superfamily.The mutant FecB of bone morphogenetic protein receptor 1B(BMPR1B)has an additive effect on ovulation rate.Six mutations(Fec^(XI),Fec^(XH),Fec^(XG),Fec^(XB),Fec^(XL),Fec^(XR))of bone morphogenetic protein 15(BMP15)related with fertility have been identified that share the same mechanism.All the mutants can increase ovulation rate in heterozygotes and cause complete sterility in homozygotes.Homozygous ewes with two new mutations(FecX^(Gr),FecX^(O))of BMP15 had increased ovulation rate without causing sterility.There are five mutations in growth differentiation factor 9(GDF9)associated with sheep prolificacy where FecG^(E) and FecG^(F) have additive an effect on ovulation rate and litter size.The newly identifiedβ-1,4-N-acetylgalactosaminyltransferase 2(B4GALNT2)gene of FecL is proposed as a new mechanism of ovulation rate regulation in sheep.Woodlands is an X-linked maternally imprinted gene which increases ovulation rate.In addition,several putative major genes need to be verified.This review is focused on the identification of the mutations and mechanisms whereby the major genes affecting ovulation rate.Qiuyue LIU Zhangyuan PAN Xiangyu WANG Wenping HU Ran DI Yaxing YAO Mingxing CHU 2014Frontiers of Agricultural Science and Engineering2014,1,4:3

18Interferon-alpha restrains growth and invasive potential of hepatocellular carcinoma induced by hepatitis B virus X protein显示文摘AIM: To investigate the effects of interferon-alpha (IFN-α) to restrain the growth and invasive potential of hepatocellular carcinoma (HCC) induced by hepatitis B virus (HBV) X protein. METHODS: The pcDNA3.1-HBx plasmid was transfected into Chang cells by Lipofectamine in vitro, and Chang/HBx was co-cultured with IFN-α. Cell survival growth curve and clonogenicity assay were used to test the growth potential of Chang/pcDNA3.1, Chang/HBx and IFN-α-Chang/HBx in vitro. Growth assay in nude mice was used to detect the growth potential of Chang/ pcDNA3.1, Chang/HBx and IFN-α-Chang/HBx in vivo. Wound healing and transwell migration assays were used to detect the invasive ability of Chang/pcDNA3.1, Chang/HBx and IFN-α-Chang/HBx. RESULTS: Compared with CCL13 cells transfected with pcDNA3.1, CCL13 with stable expression of hepatitis B virus X protein showed the characteristics of malignant cells with high capability of growth and invasion by detecting their growth curves, colony forming efficiency, wound healing , transwell migration assays and growth assays in nude mice. Its capability of growth and invasion could be controlled by IFN-α. 19Deoxygenation of methyl laurate to hydrocarbons on silica-supported Ni-Mo phosphides: Effect of calcination temperatures of precursor显示文摘SiO2-supported Ni-Mo bimetallic phosphides were prepared by temperature-programmed reduction(TPR) method from the phosphate precursors calcined at different temperatures. Their properties were characterized by means of ultraviolet-visible diffuse reflectance spectroscopy(UV-Vis DRS), H2temperature-programmed reduction(H2-TPR), X-ray diffraction(XRD), transmission electron microscopy(TEM), CO chemisorption, H2 and NH3temperature-programmed desorptions(H2-TPD and NH3-TPD). Their catalytic performances for the deoxygenation of methyl laurate were tested in a fixed-bed reactor. When the precursors were calcined at 400 and 500?C, respectively, Ni Mo P2 phase could be formed apart from Ni2 P and Mo P phases in the prepared C400 and C500 catalysts. However, when the precursors were calcined at600, 700 and 800°C, respectively, only Ni2 P and Mo P phases could be detected in the prepared C600, C700 and C800 catalysts. Also, in C400, C500 and C600 catalysts, Mo atoms were found to be entered in the lattice of Ni2 P phase, but the entering extent became less with the increase of calcination temperature. As the calcination temperature of the precursor increased, the interaction between Ni and Mo in the prepared catalysts decreased, and the phosphide crystallite size tended to increase, subsequently leading to the decrease in the surface metal site density and the acid amount. C600 catalyst showed the highest activity among the tested ones for the deoxygenation of methyl laurate. As the calcination temperature of the precursor increased, the selectivity to C12 hydrocarbons decreased while the selectivity to C11 hydrocarbons tended to increase. This can be mainly attributed to the decreased Ni-Mo interaction and the increased phosphide particle size. 20Chitinase 3-like 1 secreted by peritumoral macrophages in esophageal squamous cell carcinoma is a favorable prognostic factor for survival显示文摘AIM To identify whether chitinase 3-like 1(CHI3 L1) serves as a suitable biomarker for the prognosis of esophageal squamous cell carcinoma(ESCC) and to analyze this protein's cellular source.METHODS An ELISA was conducted to detect the concentration of CHI3 L1 in the serum of 150 ESCC patients diagnosed between January 2001 and February 2005. The prognostic relevance of CHI3 L1 was evaluated by a Kaplan-Meier and Cox regression analysis. The immunohistochemistry was reanalyzed,and fluorescent staining was utilized to explore the cellular origins of CHI3 L1. We stimulated monocyte-derived macrophages(MDMs) with either IL-6 or the supernatant of the ESCC cell line Eca-109 and later investigated the level of CHI3 L1 by q PCR and ELISA.RESULTS The level of serum CHI3 L1 was higher in older patients(≥ 60) than in patients under the age of 60(P = 0.001). The patients with higher levels of CHI3 L1 had a significantly shorter overall survival,whereas the traditional markers,carcinoembryonic antigen and squamous cell carcinoma antigen,were less effective(P > 0.05). A multivariate Cox analysis(P = 0.001) indicated that CHI3 L1 was an independent prognostic factor for ESCC patients. Peritumoral macrophages in ESCC exhibited high levels of CHI3 L1. Interleukin-6(IL-6) and the supernatant of Eca-109 containing IL-6 stimulated MDMs to secrete CHI3 L1. The serum concentration of CHI3 L1 in the ESCC patients showed a weak correlation with the laboratory inflammatory parameters neutrophil(NEU,P = 0.045),neutrophil/lymphocyte rate(NLR,P = 0.016),and C-reactive protein(CRP,P < 0.001).CONCLUSION Our study first established a connection between the pretreated CHI3 L1 and patients with ESCC,and the serum CHI3 L1 was primarily secreted by ESCC-surrounded macrophages.Shan Xing Xin Zheng Tao Zeng Mu-Sheng Zeng Qian Zhong Yue-Song Cao Kai-Lu Pan Chu Wei Fan Hou Wan-Li Liu 2017World Journal of Gastroenterology2017,23,43:2
