1Source mechanism of strong aftershocks (M_s≥5.6) of the 2008/05/12 Wenchuan earthquake and the implication for seismotectonics显示文摘Dozens of >M5, hundreds of >M4, and much more >M3 aftershocks occurred after the 2008/05/12 Wenchuan earthquake, which were well recorded by permanent and portable seismic stations. After relocated with P arrival, the >M3 aftershocks show two trends of distribution, with most of the aftershocks located along the north-east strike consistent with Longmenshan fault system, yet there is a north-west trend around the epicenter. It seems that substantially more aftershocks occur in regions with crystalline bedrocks. Then we collected waveform data from National Digital Seismograph Network and regional seismograph network of China, and employed 'Cut and Paste' method to obtain focal mechanisms and depths of the big aftershocks (M≥5.6). While most of those aftershocks show thrust mechanism, there are some strike slip earthquakes in the northern-most end of the rupture. Focal mechanisms show that the events located on the southern part of central Beichuan-Yingxiu Fault (BY) are mainly thrust earthquakes, which is consistent with initial mechanism of the main shock rupture. In the north part the aftershocks along the BY are also dominated by thrust slip, which is quite different from the right slip rupture of the main shock. Around Qingchuan-Pingwu Fault, the focal mechanisms are dominated by right-slip rupture with large depths (~18 km). So we suspected that in the north part the main shock might rupture on two faults: Beichuan Fault and Qingchuan-Pingwu Fault. 2Effect of phosphorylation of MAPK and Stat3 and expression of c-fos and c-jun proteins on hepatocarcinogenesis and their clinical significance显示文摘AIM To study the effect of phosphorylation ofMAPK and Stat3 and the expression of c-fos andc-jun proteins on hepatocellular carcinogenesisand their clinical significance.METHODS SP immunohistochemistry was usedto detect the expression of p42/44MAPK, p-Stat3,c-fos and c-jun proteins in 55 hepatocellularcarcinomas (HCC) and their surrounding livertissues.RESULTS The positive rates and expressionlevels of p42/44MAPK, p-Stat3, c-fos and c-junproteins in HCCs were significantly higher thanthose in pericarcinomatous liver tissues (PCLT).A positive correlation was observed between theexpression of p42/44MAPK and c-fos proteins, andbetween p-Stat3 and c-jun, but there was nosignificant correlation between p42/44MAPK and p-Stat3 in HCCs and their surrounding livertissues.CONCLUSION The abnormalities of Ras/Rat/MAPK and JAKs/ Stat3 cascade reaction maycontribute to malignant transformation ofhepatocytes. 3Expression of vascular endothelial growth factor and its role in oncogenesis of human gastric carcinoma显示文摘AIM To establish the role of vascular endothelial growth factor (VEGF) in the oncogenesisof human gastric carcinoma more directly.METHODS The expression of VEGF and its receptor kinase-domain insert containing receptor (KDR) in human gastric cancer tissue were observed by immunohistochemical staining. VEGF levels were manipulated in human gastric cancer cell using eukaryotic expression constructs designed to express the complete VEGF165 complimentary DNA in either the sense or antisense orientation. The biological changes of the cells were observed in which VEGF was up-regulated or downregulated.RESULTS VEGF-positive rate was 50%, and VEGF was mainly localized in the cytoplasm and membrane of the tumor cells, while KDR was mainly located in the membrane of vascular endothelial cells in gastric cancer tissues and peri-cancerous tissue. In 2 cases of 50 specimens, the gastric cancer cells expressed KDR,localized in both the cytoplasm and membrane.Introduction of VEGF165 antisense into human gastric cancer cells ( SGC-7901, immunofluorescence intensity,31.6%)) resulted in a significant reduction in VEGFspecific messenger RNA and total and cell surface VEGF protein ( immunofluorescence intensity, 8.9%)(P<0.05). Conversely, stable integration of VEGF165 in the sense orientation resulted in an increase in cellular and cell surface VEGF (immunofluorescence intensity,75.4%) (P<0.05). 4(-)-Epigallocatechin-3-gallate inhibits growth of gastric cancer by reducing VEGF production and angiogenesis显示文摘AIM: To investigate the effect of (-)-epigallocatechin- 3-gallate (EGCG) on growth of gastric cancer and its possible mechanism. METHODS: Heterotopic tumors were induced by subcutaneously injection of SGC-7901 cells in nude mice. Tumor growth was measured by calipers in two dimensions. Tumor angiogenesis was determined with tumor microvessel density (MVD) by immunohistology. Vascular endothelial growth factor (VEGF) protein level and activation of signal transducer and activator of transcription 3 (Stat3) were examined by Western blotting. VEGF mRNA expression was determined by RT-PCR and VEGF release in tumor culture medium by ELISA. VEGF-induced cell proliferation was studied by MTT assay, cell migration by gelatin modified Boyden chamber (Transwell) and in vitro angiogenesis by endothelial tube formation in Matrigel. RESULTS: Intraperitoneal injection of EGCG inhibited the growth of gastric cancer by 60.4%. MVD in tumor tissues treated with EGCG was markedly reduced. EGCG treatment reduced VEGF protein level in vitro and in vivo. Secretion and mRNA expression of VEGF in tumor cells were also suppressed by EGCG in a dose-dependent manner. This inhibitory effect was associated with reduced activation of Stat3, but EGCG treatment did not change the total Stat3 expression. EGCG also inhibited VEGF-induced endothelial cell proliferation, migration and tube formation. 5Prediction of recurrence and prognosis in patients with hepatocellular carcinoma after resection by use of CLIP score显示文摘AIM: The survival time of patients with hepatocellular carcinoma (HCC) after resection is hard to predict. Both residual liver function and tumor extension factors should be considered. A new scoring system has recently been proposed by the Cancer of the Liver Italian Program (CLIP).CUP score was confirmed to be one of the best ways to stage patients with HCC. To our knowledge, however, the literature concerning the correlation between CLIP score and prognosis for patients with HCC after resection was not pubhshed. The aim of this study is to evaluate the recurrence and prognostic value of CLIP score for the patients with HCC after resection.METHODS: A retrospective survey was carried out in 174patients undergoing resection of HCC from January 1986 toJune 1998. Six patients who died in the hospital afteroperation and 11 patients with the recurrence of the diseasewere excluded at 1 month after hepatectomy. By the end ofJune 2001, 4 patients were lost and 153 patients with curativeresection have been followed up for at least three years.Among 153 patients, 115 developed intrahepatic recurrenceand 10 developed extrahepatic recurrence, whereas the other28 remained free of recurrence. Recurrences were classifiedinto early ( < / = 3 year) and late ( > 3 year) recurrence. TheCLIP score included the parameters involved in the Child-Pugh stage (0-2), plus macroscopic tumor morphology (0-2), AFP levels (0-1), and the presence or absence of portalthrombosis (0-1)o By contrast, portal vein thrombosis wasdefined as the presence of tumor emboli within vascularchannel analyzed by microscopic examination in this study.Risk factors for recurrence and prognostic factors forsurvival in each group were analyzed by the chi-square test,the Kaplan-Meier estimation and the COX proportionalhazards model respectively.RESULTS: The 1-, 3-, 5-, 7-, andl0-year disease-free survivalrates after curative resection of HCC were 57.2 %、 28.3 %、23.5 %、 18.8 % and 17.8 %, respectively. Median survivaltime was 28,16,10,4,and 5 mo for CLIP score 0,1,2,3, and 4to 5, respectively. Early and late recurrence developed in109 patients and 16 patients respectively. By the chi-squaretest, tumor size, micrusatsllite, venous invasion, tumortype (uninodular, muitinodular, massive), tumor extension( < / = or > 50 % of liver parenchyna replaced by tumor),TNM stage,CLIP score,and resection margin were the risk factors for early recurrence,whereas CLIP score and Child-Pugh stage were significant risk factors for late recurrence. in univariate survival analysis,Child-Pugh stages,resection margin,tumor size,microsatellite,venous invasion,tumor type,tumor extension,TNM stages,and CLIP score were associated with prognosis.The multivariale analysis by COX proportional hazards modes showed that the independent predictive factors of survival were resection margins and TNM stages. 6Expression of p57^(kip2), Rb protein and PCNA and their relationships with clinicopathology in human pancreatic cancer显示文摘AIM: To investigate the effects of inhibiting factor of cellcycle regulation p57kip2, retinoblastinoma protein (Rb protein)and proliferating cell nuclear antigen (PCNA) in the genesisand progression of human pancreatic cancer.METHODS: The expression of p57kip2, Rb protein and PCNAin tumor tissues and adjacent tissues of 32 patients withpancreatic cancer was detected with SP immunohistochemicaltechnique.RESULTS: p57kip2 protein positive-expression rate in tumortissues of pancreatic cancer was 46.9 %, which was lowerthan that in adjacent pancreatic tissues (75.0 %) (x2=5.317,P<0.05), p57kip2 protein positive-expression correlatedsignificantly with tumor cell differentiation (well-differentiationversus moderate or low-differentiation, P<0.05) but did notcorrelate significantly with lymph node metastasis (lymph nodemetastasis versus non-lymph node metastasis, P>0.05); Rbgene protein positive-expression rate in tumor tissues was50.0 %, which was also lower than that in adjacent pancreatictissues (78.1%) (x2=5.497, P<0.05); PCNA positive-expression rate was 71.9 %, being higher than that inadjacent pancreatic tissues (43.8 %) (x2=5.189, P<0.05),PCNA positive-expression also correlated significantly withtumor cell differentiation and lymph node metastasis (well-differentiation versus moderate or low- differentiation, lymphnode metastasis versus non-lymph node metastasis, P<0.05).Rb protein positive-expression rate in the tumor tissues ofp57kip2 protein positive-expression group was 53.3 %; andRb protein positive-expression rate in the tumor tissues ofp57kip2 protein negative-expression group was 47.1%. 7Clostridium difficile infections in China显示文摘Clostridium difficile (C. difficile) infection has become one of the major hospital-associated infections in West-ern countries in the last two decades. However, there is limited information on the status of C. difficile infection inChinese healthcare settings. Given the large and increasing elderly population and the well-recognized problem ofover-prescribing of broad spectrum antibiotics in China, it is critical to understand the epidemiology and potentialrisk factors that may contribute to C. difficile infection in China. A literature review of available published studies,including those in Chinese language-based journals, was conducted. A review of the currently available literaturesuggested the presence of C. difficile infections in China, but also suggested that these infections were not particu-larly endemic. This finding should lead to better designed and greatly expanded studies to provide a more reliableepidemiologically-based conclusion on the actual status of C. difficile infection in China, including the identifica-tion of any associated risk factors. 8Detection of soluble TRAIL in HBV infected patients and its clinical implications显示文摘AIM: To detect the expression of soluble TRAIL (TNF-related apoptosis inducing ligand, TRAIL) in the peripheral blood of HBV infected patients and try to elucidate whether the expression level of sTRAIL have any correlativity with the clinical staging, the expression level of HBV markers and the degree of liver damage.METHODS: 52 cases of HBV infected patients were investigated, induding 8 HBV carriers, 30 chronic hepatitis B, 11 drrhotics and 3 HBV infection related hepatocellular carcinoma. Expression of soluble TRAIL and markers of the hepatitis B were mearsured by enzyme-linked immunosorbent assay.RESULTS: The expression level of sTRAIL in the peripheral blood of the HBV infected patients was significantly higher than that of healthy controls (1378.35±540.23 pg/ml vs 613.75±175.80 pg/ml, P<0.001). In the group of chronic hepatitis, the expression level of sTRAIL was coincident with the status of the disease and was significantly correlated with the level of ALT. In the group of cirrhosis and liver cancer, its expression level was significantly higher than that of the healthy persons and HBV carriers, but lower than that of the hepatitis B patients; meanwhile, the expression of siRAIL did not have any correlativity with the functional indexes of the liver. CONCLUSION: The soluble TRAIL in the HBV infected people may participate in the liver damage. 9The Inhibitory Effect of Extracts From Fructus lycii and Rhizoma polygonati on in vitro DNA Breakage by Alternariol显示文摘Alternariol caused DNA single-strand breakage. Conversion of the closed circular double-stranded supercoiled DNA (pBR 322) to the nicked circular form and linear form was used to investigate the effect of extracts of some Chinese medical herbs on DNA nicking induced by alternariol. Some substances in the extracts of Rhizoma polygonati (RP) and Fructus lycii (FL) were shown to protect DNA from the attack by alternariol.Some substance in the RP may bind to plasmid DNA, and this binding reduces the electrophoretic mobility of DNA. These results indicate that substances from FL and RP may be used as DNA protectors. The physiological mechanisms and genetic basis of the erect and large panicle super-high-yield plant type model for breeding japonica super rice were analyzed mainly on the panicle type, number of large vascular bundles (LVB) in the panicle neck, and the panicle type index (PTI). In the production point of view, we suggested that, for the breeding of super-high-yield japonica rice, the erect panicle types with more LVB numbers in the panicle neck and superior upper grains in the secondary branches would be the key factors. 12Embryonic hepatocyte transplantation for hepatic cirrhosis:Efficacy and mechanism of action显示文摘AIM:To investigate the efficacy and mechanism of action of allogeneic embryonic hepatocyte transplanta-tion for the treatment of hepatic cirrhosis.METHODS:Rat embryonic hepatocytes were char-acterized by examining cell markers.Wistar rats with CCl 4-induced cirrhosis were randomly divided into two groups:a model group receiving continuous CCl 4,and a cell transplantation group receiving continuous CCl 4 and transplanted with embryonic fluorescent-labeled hepatocytes.In addition,a normal control group was composed of healthy rats.All rats were sacrificed after 2 wk following the initiation of the cell transplant.Ul-trasound,pathological analyses and serum biochemical tests were used to evaluate the efficacy of embryonic hepatocyte transplantation.To analyze the recov-ery status of cirrhotic hepatocytes and the signaling pathways influenced by embryonic hepatocyte trans-plantation,real-time polymerase chain reaction was performed to examine the mRNA expression of stellate activation-associated protein (STAP),c-myb,α smooth muscle actin (α-SMA) and endothelin-1 (ET-1).West-ern blotting and immunohistochemistry were employed to detect α-SMA and ET-1 protein expression in hepatic tissues.RESULTS:Gross morphological,ultrasound and his-topathological examinations,serum biochemical tests and radioimmunoassays demonstrated that hepatic cir-rhosis was successfully established in the Wistar rats.Stem cell factor receptor (c-kit),hepatocyte growth factor receptor (c-Met),Nestin,α fetal protein,albu-min and cytokeratin19 markers were observed in the rat embryonic hepatocytes.Following embryonic hepa-tocyte transplantation,there was a significant reversal in the gross appearance,ultrasound findings,histo-pathological properties,and serum biochemical param-eters of the rat liver.In addition,after the activation of hepatic stellate cells and STAP signaling,α-SMA,c-myb and ET-1 mRNA levels became significantly lower than in the untreated cirrhotic group (P < 0.05).These levels,however,were not statistically different from those of the normal healthy group.Immunohisto-chemical staining and Western blot analyses revealed that α-SMA and ET-1 protein expression levels in the transplantation group were significantly lower than in the untreated cirrhotic group,but being not statistically different from the normal group.CONCLUSION:Transplantation of embryonic hepato-cytes in rats has therapeutic effects on cirrhosis.The described treatment may significantly reduce the ex-pression of STAP and ET-1.Wen-Ting Bin,Qing Xu,Xiao-Lin Shi,Laboratory of Reproductive Biology,Department of Histology and Embryology,School of Basic Medical Sciences,Capital Medical University,Beijing 100069,China Li-Mei Ma,Department of Histology and Embryology,Kunming Medical University,Kunming 650031,Yunnan Province,China 2012World Journal of Gastroenterology2012,18,4:4
13Interaction of host-guest complexes of cucurbit[n]urils with double probe guests显示文摘Three guests with two moiety probes for different Cucurbit[n = 6—8]urils have been synthesized. They are N-(2-methylenethiophen)-adamataneamine, N-(2-methylene pyrrole)- adamataneamine and N-(2-methylenefurfuran)-adamataneamine. The probes are methyle-nepyridyl typically for Q[6] and adamataneamine typically for Q[7]. The host-guest complexes of Cucurbit[n = 6—8]urils with these guests have been investigated by using NMR techniques and ESMS method. 15Lowered HGK expression inhibits cell invasion and adhesion in hepatocellular carcinoma cell line HepG2显示文摘AIM:To investigate the effects of RNA interference tar-geting hepatocyte progenitor kinase-like kinase(HGK) in the invasion and adhesion of hepatocellular carcinoma(HCC) cell line HepG2.METHODS:Three paired insert DNA fragments specif ic to HGK gene and one negative control DNA fragment were synthesized and inserted into RNAi-Ready pSIREN-RetroQ-ZsGreen vector.Western blotting assay and real-time reverse transcriptase polymerase chain reaction(RT-PCR) were used to screen the vector with a highest inhibitory rate.The vector was used to generate recom-binant retrovirus specif ic to HGK.3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl-2h-tetrazolium bromide(MTT) assay was used to examine cell growth;wound closure assay and cell adhesion assay were employed to investigate cell migration and adhesion respectively;and transwell assay and three-dimensional culture invasion assay were used to detect cell invasion.The expressions of matrix metalloproteinase(MMP)-2,MMP-9 and nuclear factor(NF)-κB were detected by Western blotting assay.RESULTS:The real time RT-PCR and Western blotting assay showed that cells transfected with retrovirus me-diating RNAi targeting of HGK(RV-shHGK)-1 vector had the strongest inhibition of HGK protein,with an inhibi-tion rate of 76%,and this vector was used to generate recombinant retrovirus RV-shHGK-1.Cell adhesion assay and MTT assay found that cell adhesion and growth of the cells infected with RV-shHGK-1 were significantly lower than those of the control cells(P < 0.05).Wound closure assay,transwell assay and three-dimensional culture invasion assay showed that the cell invasiveness was significantly less in HGK knockdown cells than in the control cells(P < 0.05).The expressions of MMP-2,MMP-9 and NF-κB were inhibited in HepG2 cells infected with RV-shHGK-1.CONCLUSION:Down-regulation of HGK can obviously inhibit the migration and invasion of HepG2 cells in vitro.HGK may be a new therapeutic target for treatment of HCC.Su-Xia Han,Qing Zhu,Jin-Lu Ma,Jing Zhao,Xi Jia,Dan Zhang,Oncology Center of the First Aff iliated Hospital,College of Medicine,Xi’an Jiaotong University,Xi’an 710061,Shaanxi Province,ChinaChen Huang,Central Laboratory,College of Medicine,Xi’an Jiaotong University,Xi’an 710061,Shaanxi Province,China 2010World Journal of Gastroenterology2010,16,36:3
16A mutation specific polymerase chain reaction for detecting hepatitis B virus genome mutations at nt551显示文摘AIM: The hepatitis B surface antigen (HBsAg) is considered to be one of the best markers for the diagnosis of acute and chronic HBV infection. But in some patients, this antigen cannot be detected by routine serological assays despite the presence of virus. One of the most important explanations for the lack of detectable HBsAg is that mutations which occur within the 'a' determinant of HBV S gene can alter expression of HBsAg and lead to changes of antigenicity and immunogenicity of HBsAg accordingly. As a result, these mutants cannot be detected by diagnosis assays. Thus, it is essential to find out specific and sensitive methods to test the new mutants and further investigate their distribution. This study is to establish a method to investigate the distribution of the HBsAg mutant at nt551.METHODS: A mutation specific polymerase chain reaction (msPCR) was established for amplifying HBV DNA with a mutation at nt551. Four sets of primer pairs, P551A-PPS,P551G-PPS, P551C-PPS and P551T-PPS, with the same sequences except for one base at 3' terminus were designed and synthesized according to the known HBV genome sequences and the popular HBV subtypes, adr and adw, in China. At the basis of regular PCR method, we explored the specific conditions for amplifying HBV DNAs with a mutation at nt551 by regulating annealing temperature and the concentration of these primers. 126 serum samples from patients of hepatitis B were collected, among which 16 were positive for HBV S DNA in the nested PCR amplification. These 16 HBV S DNAs were detected by using the msPCR method.RESULTS: When the annealing temperature was raised to 71 °C, nt551A and nt551G were amplified specifically by P551A-PPS and P551G-PPS; At 72 °C and 5 pmole of the primers (each) in reaction of 25 μl volume, nt551C and nt551T were amplified specifically by P551C-PPS and P551TPPS. 16 of HBV S gene fragments were characterized by using this method. 14 of them were positive for nt551A, one was positive for nt551G, and the other one was positive for nt551T. 17Direct in vivo injection of ^(131)I-GMS and its distribution and excretion in rabbit显示文摘AIM: To explore the distribution and metabolism of 131 I-gelatin microspheres ( 131 I-GMSs) in rabbits after direct injection into rabbits’ livers.METHODS: Twenty-eight healthy New Zealand rabbits were divided into seven groups,with four rabbits per group.Each rabbit’s hepatic lobes were directly injected with 41.336 ± 5.106 MBq 131 I-GMSs.Each day after 131 I-GMSs administration,4 rabbits were randomly selected,and 250 μL of serum was collected for γ count.Hepatic and thyroid functions were tested on days 1,4,8,16,24,32,48 and 64 after 131 I-GMSs administration.Single-photon emission computed tomography (SPECT) was taken for each group on days 0,1,4,8,16,24,32,48,64 after 131 I-GMSs administration.A group of rabbits were sacrificed respectively on days 1,4,16,24,32,48,64 after 131 I-GMSs administration.Their livers were taken out for histological examination.RESULTS: After 131 I-GMSs administration,the nuclide was collected in the hepatic area with microspheres.The radiation could be detected on day 48 after 131 IGMSs administration,and radiography could be seen in thyroid areas in SPECT on days 4,8,16 and 24.One day after 131 I-GMSs administration,the liver function was damaged but recovered 4 d later.Eight days after 131 I-GMSs administration,the levels of free triiodothyronine and free thyroxin were reduced,which restored to normal levels on day 16.Histological examination showed that the microspheres were degraded to different degrees at 24,32 and 48 d after 131 I-GMSs administration.The surrounding parts of injection points were in fibrous sheathing.No microspheres were detected in histological examination on day 64 after 131 I-GMSs administration.CONCLUSION: Direct in vivo injection of 131 I-GMSs is safe in rabbits.It may be a promising method for treatment of malignant tumors.Yu Ma,Yi Wan,Li-Geng Duan,Xiao-Li Chen,The Research Unit of Hepato-Bilio-Pancreatology and Department of Hepatic Surgery,West China Hospital of Sichuan University,Chengdu 610041,Sichuan Province,China Dong-Hui Luo,Chuan-Qin Xia,College of Chemistry,Sichuan University,Chengdu 610041,Sichuan Province,China Lin Li,Department of Nuclear Medicine and The National Key Discipline of Medical Imaging and Nuclear Medicine,West China Hospital of Sichuan University,Chengdu 610041,Sichuan Province,China 2010World Journal of Gastroenterology2010,16,17:3

19Discovery of Early Triassic conodonts in western Gangdisê and the establishment of the Tangnale Formation显示文摘Most geologists believe that there are no Early and Middle Triassic strata in the W. Gandisê stratigraphic subregion, but the present authors have found Early Triassic conodonts for the first time in the Shiquanhe area, including five conodonts genera (Form genera): Pachycladina, Neohindeodella, Cornudina, Hadrodontina and Hibbardella sp. etc. Then we affirm that Early Triassic deposits exist in the Gandisê stratigraphic subregion, and establish the Tangnale Formation.
