1Gene cloning and expression of cadherin in midgut of Helicoverpa armigera and its Cry1A binding region显示文摘Cadherins belong to one of the families of animal glycoproteins responsible for cal-cium-dependent cell-cell adhesion. Recent literatures showed that the cadherin-like in midgut of several insects served as the receptor of Bt toxin Cry1A and the variation of cadherin-like is re-lated to insect’s resistance to Cry1A. The full-length cDNA encoding cadherin-like of Helicoverpa armigera is cloned by degenerate PCR and RACE techniques and the gene was designated as BtR-harm, which is 5581 bp in full-length, encoding 1730 amino acid residues (BtR-harm was deposited in GenBank and the accession number is AF519180). Its predicted molecular weight and isoelectric point were 195.39 kDa and 4.23, respectively. The inferred amino acid sequence includes a signal sequence, 11 cadherin repeats, a membrane-proximal region, a transmem-brane region and a cytoplasmic region. Sequence analysis indicated that the deduced protein sequence was most similar to the cadherin-like from Heliothis virescens with 84.2% identity and highly similar to three other lepidopteran cadherin from Bombyx mori, Manduca sexta and Pectinophora gossypiella, with the sequence identities of 60.3.6%, 57.5% and 51.0%, respec-tively. The cDNA encoding cadherin gene was expressed successfully in E. coli and the recom-binant proteins can bind with Cry1Ac. Truncation analysis and binding experiment of BtR-harm revealed that the Cry1A binding region was a contiguous 244-amino acid sequence, which lo-cated between amino acid 1217 and 1461. Semi-quantitative RT-PCR analysis showed that BtR-harm was highly expressed in midgut of H. armigera, very low expressed in foregut and hindgut and was not expressed in other tissues. After H. armigera producing resistance to Cry1Ac, the expression quantity of BtR-harm significantly decreased in midgut of H. armigera. It is the first confirmation that BtR-harm can function as receptor of Cry1Ac in H. armigera and the binding region was located on a contiguous 244 amino acid sequence, suggesting that the de-crease of expression quantity of BtR-harm is one of the main reasons for H. armigera resistance to Cry1Ac.WANG Guirong WU Kongming LIANG Gemei GUO Yuyuan 2005Science China(Life Sciences)2005,48,4:17
2Notch signaling pathway:architecture,disease,and therapeutics显示文摘The NOTCH gene was identified approximately 110 years ago.Classical studies have revealed that NOTCH signaling is an evolutionarily conserved pathway.NOTCH receptors undergo three cleavages and translocate into the nucleus to regulate the transcription of target genes.NOTCH signaling deeply participates in the development and homeostasis of multiple tissues and organs,the aberration of which results in cancerous and noncancerous diseases.However,recent studies indicate that the outcomes of NOTCH signaling are changeable and highly dependent on context.In terms of cancers,NOTCH signaling can both promote and inhibit tumor development in various types of cancer.The overall performance of NOTCH-targeted therapies in clinical trials has failed to meet expectations.Additionally,NOTCH mutation has been proposed as a predictive biomarker for immune checkpoint blockade therapy in many cancers.Collectively,the NOTCH pathway needs to be integrally assessed with new perspectives to inspire discoveries and applications.In this review,we focus on both classical and the latest findings related to NOTCH signaling to illustrate the history,architecture,regulatory mechanisms,contributions to physiological development,related diseases,and therapeutic applications of the NOTCH pathway.The contributions of NOTCH signaling to the tumor immune microenvironment and cancer immunotherapy are also highlighted.We hope this review will help not only beginners but also experts to systematically and thoroughly understand the NOTCH signaling pathway.Binghan Zhou Wanling Lin Yaling Long Yunkai Yang Huan Zhang Kongming Wu Qian Chu 2022Signal Transduction and Targeted Therapy2022,7,4:14
3Cotton bollworm resistance to Bt transgenic cotton: A case analysis显示文摘Cotton bollworm (Helicoverpa armigera) is one of the most serious insect pests of cotton. Transgenic cotton expressing Cry toxins derived from a soil bacterium, Bacillus thuringiensis (Bt), has been produced to target this pest. Bt cotton has been widely planted around the world, and this has resulted in efficient control of bollworm populations with reduced use of synthetic insecticides. However, evolution of resistance by this pest threatens the continued success of Bt cotton. To date, no field populations of bollworm have evolved significant levels of resistance; however, several laboratory-selected Cry-resistant strains of H. armigera have been obtained, which suggests that bollworm has the capacity to evolve resistance to Bt. The development of resistance to Bt is of great concern, and there is a vast body of research in this area aimed at ensuring the continued success of Bt cotton. Here, we review studies on the evolution of Bt resistance in H. armigera, focusing on the biochemical and molecular basis of Bt resistance. 4Cloning,expression and binding specificity analysis of odorant binding protein 3 of the lucerne plant bug,Adelphocoris lineolatus(Goeze)显示文摘Credible evidence shows that odorant binding proteins(OBPs)are required for insect olfaction perception and play a key role in transporting hydrophobic odorants across the sensillum lymph to the olfactory receptors(ORs).In the present study,a novel OBP(AlinOBP3)gene from the lucerne plant bug,Adelphocoris lineolatus,was cloned and expressed.The expression pattern of AlinOBP3 was evaluated by qPCR,which indicated that AlinOBP3 was dominantly expressed in antennae.The binding properties of AlinOBP3 with 9 cotton volatiles and 5 sex pheromone analogs were measured by fluorescence competitive binding assays with the fluorescence probe 1-NPN.The results revealed that of 9 cotton volatiles,Myrcene,β-Ocimene and α-Phellandrene can bind with AlinOBP3.α-Phellandrene especially bound to AlinOBP3 with a high binding affinity,with a dissociation constant of 56.68 μmol/L.Of the 5 sex pheromone analogs,Hexyl butyrate had the strongest binding affinity with AlinOBP3,with a dissociation constant as 59.53 μmol/L.Butyl butyrate,trans-2-Hexenyl butyrate and Ethyl butyrate had medium binding affinities with AlinOBP3,with dissociation constants of 227.39,108.77 and 143.47 μmol/L,respectively.The results suggest that AlinOBP3 might be a pheromone binding protein(PBP)with a dual-function for the perception of sex pheromones and plant volatiles.GU ShaoHua SUN Yang REN LiYan ZHANG XueYing ZHANG YongJun WU KongMing GUO YuYuan 2010Chinese Science Bulletin2010,55,34:7

6The evolution of China’s regulation of agricultural biotechnology显示文摘To ensure safe use of genetically modified organisms(GMOs),since 1993,China has made great efforts to establish and improve the safety regulatory system for GMOs.Here,we summarize and analyze the regulatory framework of agricultural GMOs,and the progress in regulatory approval of GM crops in China.In general,the development of GMO safety regulations underwent four stages:exploration(1993–2000),development(2001–2010),improvement(2011–2020)and current(2021-present)stage.The first formal regulation was promulgated in 1993,which provided a basis for further development of the regulations,during the exploration stage,when insect-resistant GM cotton,expressing genes from Bacillus thuringiensis(Bt),was approved for cultivation.During the development stage,the Chinese government issued a series of administrative measures,which covered almost all the fields relative to GMO safety when the basic regulatory system was established.Along with the controversy over GMO safety,the regulations have been further,and greatly improved,during improvement stage.From 2021,a few additional revisions have been made,and meanwhile,the new regulation on gene-edited crops was introduced with the development of biotechnology,forming a relative complete regulation and law system for China.The well-developed GMO regulations establishes a firm basis for safe use of GM crops in China.Currently,GM cotton and GM papaya have been widely grown on a large scale in China that have brought great economic and ecological benefits.In addition,12 corn events,3 soybean events,and 2 rice events have also obtained biosafety certification,but presently,these lines have yet to enter commercial production.However,several GM soybean and corn events have entered pilot industrialization,and can soon be expected to be commercially grown in China.In addition to planting,six GM crops,including soybean,corn,cotton,canola,papaya and sugar beet,with a total of 64 events,have been approved for import as processing material in China.Jingang Liang Xiaowei Yang Yue Jiao Danxia Wang Qiang Zhao Yu Sun Yunhe Li Kongming Wu 2022aBIOTECH2022,3,4:2
7RESEARCH AND APPLICATION OF CROP PEST MONITORING AND EARLY WARNING TECHNOLOGY IN CHINA显示文摘The importance of food security,especially in combating the problem of acute hunger,has been underscored as a key component of sustainable development.Considering the major challenge of rapidly increasing demands for both food security and safety,the management and control of major pests is urged to secure supplies of major agricultural products.However,owing to global climate change,biological invasion(e.g.,fall armyworm),decreasing agricultural biodiversity,and other factors,a wide range of crop pest outbreaks are becoming more frequent and serious,making China,one of the world’s largest country in terms of agricultural production,one of the primary victims of crop yield loss and the largest pesticide consumer in the world.Nevertheless,the use of science and technology in monitoring and early warning of major crop pests provides better pest management and acts as a fundamental part of an integrated plant protection strategy to achieve the goal of sustainable development of agriculture.This review summarizes the most fundamental information on pest monitoring and early warning in China by documenting the developmental history of research and application,Chinese laws and regulations related to plant protection,and the National Monitoring and Early Warning System,with the purpose of presenting the Chinese model as an example of how to promote regional management of crop pests,especially of cross border pests such as fall armyworm and locust,by international cooperation across pest-related countries.Qiulin WU Juan ZENG Kongming WU 2022Frontiers of Agricultural Science and Engineering2022,9,1:2

9MAT1 correlates with molecular subtypes and predicts poor survival in breast cancer显示文摘Objective:Menage a trois 1(MAT1)is a targeting subunit of cyclin-dependent kinase-activating kinase and general transcription factor IIH kinase,which modulates cell cycle,transcription and DNA repair.Its dysregulation is responsible for diseases including cancers.To further explore the role of MAT1 in breast cancer,we investigated the pathways in which MAT1 might be involved,the association between MAT1 and molecular subtypes,and the role of MAT1 in clinical outcomes of breast cancer patients.Methods:We conducted immunohistochemistry staining on tissue microarray and immunofluorescence staining on sections of MAT1 stable breast cancer cells.Also,we performed Kyoto Encyclopedia of Genes and Genomes pathway analysis,correlation analysis and prognosis analysis on public databases.Results:MAT1 was involved in multiple pathways including normal physiology signaling and disease-related signaling.Furthermore,MAT1 positively correlated with the protein status of estrogen receptor and progesterone receptor,and was enriched in luminal-type and human epidermal growth factor receptor 2-enriched breast cancer in comparison with basal-like subtype at both m RNA and protein levels.Correlation analysis revealed significant association between MAT1 m RNA amount and epithelial markers,mesenchymal markers,cancer stem cell markers,apoptosis markers,transcription markers and oncogenes.Consistently,the results of immunofluorescence stain indicated that MAT1 overexpression enhanced the protein abundance of epidermal growth factor receptor,vimentin,sex determining region Y-box 2 and sine oculis homeobox homolog 1.Importantly,Kaplan-Meier Plotter analysis reflected that MAT1 could serve as a prognostic biomarker predicting worse relapse-free survival and metastasis-free survival.Conclusions:MAT1 is correlated with molecular subtypes and is associated with unfavorable prognosis for breast cancer patients.Hanxiao Xu Xianguang Bai Shengnan Yu Qian Liu Richard G Pestell Kongming Wu 2018Chinese Journal of Cancer Research2018,30,3:2
