Relationship between Cognition Function and Hippocampus Structure after Long-term Microwave Exposure显示文摘Objective To analyze the effects of long-term microwave exposure on hippocampal structure and function in the rat. Methods Experiments were performed on 184 male Wistar rats(three exposure groups and a sham group).Microwaves were applied daily for 6 min over 1 month at average power densities of 2.5,5,and 10 mW/cm2.Learning and memory abilities were assessed by Morris water maze.High performance liquid chromatography was used to detect neurotransmitter concentrations in the hippocampus.Hippocampal structures were observed by histopathological analysis. Results Following long-term microwave exposure there was a significant decrease in learning and memory activity in the 7 d,14 d,and 1 m in all three microwave exposure groups.Neurotransmitter concentrations of four amino acids(glutamate, aspartic acid, glycine, and gamma-aminobutyric acid) in hippocampus were increased in the 2.5 and 5 mW/cm2 groups and decreased in the 10 mW/cm2 group. There was evidence of neuronal degeneration and enlarged perivascular spaces in the hippocampus in the microwave exposure groups.Further, mitochondria became swollen and cristae were disordered. The rough endoplasmic reticulum exhibited sacculated distension and there was a decrease in the quantity of synaptic vesicles. Conclusion These data suggest that the hippocampus can be injured by long-term microwave exposure, which might result in impairment of cognitive function due to neurotransmitter disruption.ZHAO Li PENG Rui Yun WANG Shui Ming WANG Li Feng GAO Ya Bing DONG Ji LI Xiang SU Zhen Tao 2012Biomedical and Environmental Sciences2012,25,2:27
Echinococcus Granulosus 14-3-3 Protein:A Potential Vaccine Candidate Against Challenge with Echinococcus Granulosus in Mice显示文摘Objective To investigate the protective immunity against Echinococcus granulosus in mice immunized with rEg14-3-3.Methods ICR mice were subcutaneously immunized three times with rEg14-3-3,followed by the challenge with Echinococcus granulosus protoscoleces intraperitoneally and then sacrificed after six months of post-challenge to detect the proliferation of splenocytes by MTT assay, and to measure the secretion of IL-2,IL-4,IL-10,and IFN-γ by ELISA.The rate of reduced hydatid cyst and the levels of IgE,IgG and IgG subclasses in sera were examined. Results Mice vaccinated with rEg14-3-3 and challenged with protoscoleces revealed significant protective immunity of 84.47%.ELISA analysis indicated that the immunized mice generated specific high levels of IgG and the prevailing isotypes of IgG were IgG1 and IgG2a.Splenocytes from mice immunized with rEg14-3-3 showed a significant proliferation response. The secretion of IFN-γ and IL-2 increased significantly in the vaccinated mice whereas there was no significant difference in IL-4 and IL-10 levels between vaccinated and control mice. Conclusion The results indicate that the rEg14-3-3 vaccine could induce a high level of protective immunity as a promising vaccine candidate to prevent cystic echinococcosis.LI Zong Ji WANG Ya Na WANG Qi ZHAO Wei 2012Biomedical and Environmental Sciences2012,25,3:18
Effectiveness of Different Waist Circumference Cut-off Values in Predicting Metabolic Syndrome Prevalence and Risk Factors in Adults in China显示文摘Objective To study the effectiveness of waist circumference cut-off values in predicting the prevalence of metabolic syndrome(MetS) and risk factors in adults in China. Methods A cross-sectional survey was condcuted in 14 provinces(autonomous region, municipality) in China. A total of 47 325 adults aged ≥20 years were selected by multistage stratified sampling, and questionnaire survey and physical and clinical examination were conducted among them. MetS was defined according to the International Diabetes Federation(IDF) criteria and modified IDF criteria. Results The age-standardized prevalence of MetS was 24.2%(22.1% in men and 25.8% in women) and 19.5%(22.1% in men and 18.0% in women) according to the IDF criteria and modified IDF criteria respectively. The age-standardized prevalence of pre-MetS was 8.1%(8.6% in men and 7.8% in women) according to the modified IDF criteria. The prevalence of MetS was higher in urban residents than rural residents and in northern China residents than in southern China residents. The prevalence of central obesity was about 30% in both men and women according to the ethnicity-specific cut-off values of waist circumference for central obesity(90 cm for men and 85 cm for women). Multivariate regression analysis revealed no significant difference in risk factors between the two MetS definitions. Conclusion Using both the modified IDF criteria and ethnicity-specific cut-off values of waist circumference can provide more useful information about the prevalence of MetS in China.ZHOU Hai Cheng LAI Ya Xin SHAN Zhong Yan JIA Wei Ping YANG Wen Ying LU Ju Ming WENG Jian Ping JI Li Nong LIU Jie TIAN Hao Ming JI Qiu He ZHU Da Long CHEN Li GUO Xiao Hui ZHAO Zhi Gang Li Qiang ZHOU Zhi Guang GE Jia Pu SHAN Guang Liang 2014Biomedical and Environmental Sciences2014,27,5:17
Microwave Exposure Impairs Synaptic Plasticity in the Rat Hippocampus and PC12 Cells through Over-activation of the NMDA Receptor Signaling Pathway显示文摘Objective The aim of this study is to investigate whether microwave exposure would affect the N-methyl-D-aspartate receptor(NMDAR) signaling pathway to establish whether this plays a role in synaptic plasticity impairment. Methods 48 male Wistar rats were exposed to 30 m W/cm2 microwave for 10 min every other day for three times. Hippocampal structure was observed through H&E staining and transmission electron microscope. PC12 cells were exposed to 30 m W/cm2 microwave for 5 min and the synapse morphology was visualized with scanning electron microscope and atomic force microscope. The release of amino acid neurotransmitters and calcium influx were detected. The expressions of several key NMDAR signaling molecules were evaluated. Results Microwave exposure caused injury in rat hippocampal structure and PC12 cells, especially the structure and quantity of synapses. The ratio of glutamic acid and gamma-aminobutyric acid neurotransmitters was increased and the intracellular calcium level was elevated in PC12 cells. A significant change in NMDAR subunits(NR1, NR2 A, and NR2B) and related signaling molecules(Ca2+/calmodulin-dependent kinase II gamma and phosphorylated c AMP-response element binding protein) were examined. Conclusion 30 m W/cm2 microwave exposure resulted in alterations of synaptic structure, amino acid neurotransmitter release and calcium influx. NMDAR signaling molecules were closely associated with impaired synaptic plasticity.XIONG Lu SUN Cheng Feng ZHANG Jing GAO Ya Bing WANG Li Feng ZUO Hong Yan WANG Shui Ming ZHOU Hong Mei XU Xin Ping DONG Ji YAO Bin Wei ZHAO Li PENG Rui Yun 2015Biomedical and Environmental Sciences2015,28,1:16
Prognostic values of the integrated model incorporating the volume of metastatic regional cervical lymph node and pretreatment serum Epstein-Barr virus DNA copy number in predicting distant metastasis in patients with N1 nasopharyngeal carcinoma显示文摘Background: According to the 7 th edition of the American Joint Committee on Cancer(AJCC) staging system, over50% of patients with nasopharyngeal carcinoma(NPC) have N1 disease at initial diagnosis. However, patients with N1 NPC are relatively under-researched, and the metastasis risk of this group is not well-stratified. This study aimed to evaluate the prognostic values of gross tumor volume of metastatic regional lymph node(GTVnd) and pretreatment serum copy number of Epstein-Barr virus(EBV) DNA in predicting distant metastasis of patients with N1 NPC, and to develop an integrated prognostic model that incorporates GTVnd and EBV DNA copy number for this group of patients. Methods: The medical records of 787 newly diagnosed patients with nonmetastatic, histologically proven N1 NPC who were treated at Sun Yat-sen University Cancer Center between November 2009 and February 2012 were analyzed. Computed tomography-derived GTVnd was measured using the summation-of-area technique. Blood samples were collected before treatment to quantify plasma EBV DNA. The receiver operating characteristic(ROC) curve analysis was used to evaluate the cut-off point for GTVnd, and the area under the ROC curve was used to assess the predicted validity of GTVnd. The survival rates were assessed by Kaplan-Meier analysis, and the survival curves were compared using a log-rank test. Multivariate analysis was conducted using the Cox proportional hazard regression model. Results: The 5-year distant metastasis-free survival(DMFS) rates for patients with GTVnd > 18.9 vs.≤ 18.9 mL were82.2% vs. 93.2%(P < 0.001), and for patients with EBV DNA copy number > 4000 vs. < 4000 copies/mL were 83.5% vs.93.9%(P < 0.001). After adjusting for GTVnd, EBV DNA copy number, and T category in the Cox regression model, both GTVnd > 18.9 mL and EBV DNA copy number > 4000 copies/mL were significantly associated with poor prognosis(both P < 0.05). According to combination of GTVnd and EBV DNA copy number, all patients were divided into low-,moderate-, and high-risk groups, with the 5-year DMFS rates of 96.1,87.4, and 73.8%, respectively(P < 0.001). Multivariate analysis confirmed the prognostic value of this model for distant metastatic risk stratification(hazard ratio [HR],4.17; 95% confidence interval [CI] 2.34-7.59; P < 0.001).Conclusions: GTVnd and serum EBV DNA copy number are independent prognostic factors for predicting distant metastasis in NPC patients with N1 disease. The prognostic model incorporating GTVnd and EBV DNA copy number may improve metastatic risk stratification for this group of patients.Ji‑Jin Yao Guan‑Qun Zhou Ya‑Qin Wang Si‑Yang Wang Wang‑Jian Zhang Ya‑Nan Jin Fan Zhang Li Li Li‑Zhi Liu Zhi‑Bin Cheng Jun Ma Zhen‑Yu Qi 2017Chinese Journal of Cancer2017,36,12:9

TNF-αTNFR1 Signaling is Required for the Full Expression of Acute and Chronic Itch in Mice via Peripheral and Central Mechanisms显示文摘Increasing evidence suggests that cytokines and chemokines play crucial roles in chronic itch. In the present study, we evaluated the roles of tumor necrosis factor-alpha(TNF-a) and its receptors TNF receptor subtype-1(TNFR1) and TNFR2 in acute and chronic itch in mice.Compared to wild-type(WT) mice, TNFR1-knockout(TNFR1-KO) and TNFR1/R2 double-KO(DKO), but not TNFR2-KO mice, exhibited reduced acute itch induced by compound 48/80 and chloroquine(CQ). Application of the TNF-synthesis inhibitor thalidomide and the TNF-αantagonist etanercept dose-dependently suppressed acute itch. Intradermal injection of TNF-α was not sufficient to evoke scratching, but potentiated itch induced by compound 48/80, but not CQ. In addition, compound 48/80 induced TNF-α mRNA expression in the skin, while CQ induced its expression in the dorsal root ganglia(DRG) and spinal cord. Furthermore, chronic itch induced by dry skin was reduced by administration of thalidomide and etanercept and in TNFR1/R2 DKO mice. Dry skin induced TNF-α expression in the skin, DRG, and spinal cord and TNFR1 expression only in the spinal cord. Thus, our findings suggest that TNF-α/TNFR1 signaling is required for the full expression of acute and chronic itch via peripheral and central mechanisms, and targeting TNFR1 may be beneficial for chronic itch treatment.Xiuhua Miao Ya Huang Teng-Teng Liu Ran Guo Bing Wang Xue-Long Wang Li-Hua Chen Yan Zhou Ru-Rong Ji Tong Liu 2018Neuroscience Bulletin2018,34,1:8
Association of Overlapped and Un-overlapped Comorbidities with COVID-19 Severity and Treatment Outcomes: A Retrospective Cohort Study from Nine Provinces in China显示文摘Objective Several COVID-19 patients have overlapping comorbidities. The independent role of each component contributing to the risk of COVID-19 is unknown, and how some non-cardiometabolic comorbidities affect the risk of COVID-19 remains unclear. Methods A retrospective follow-up design was adopted. A total of 1,160 laboratory-confirmed patients were enrolled from nine provinces in China. Data on comorbidities were obtained from the patients’ medical records. Multivariable logistic regression models were used to estimate the odds ratio(OR) and 95% confidence interval(95% CI) of the associations between comorbidities(cardiometabolic or non-cardiometabolic diseases), clinical severity, and treatment outcomes of COVID-19.Results Overall, 158(13.6%) patients were diagnosed with severe illness and 32(2.7%) had unfavorable outcomes. Hypertension(2.87, 1.30–6.32), type 2 diabetes(T2 DM)(3.57, 2.32–5.49),cardiovascular disease(CVD)(3.78, 1.81–7.89), fatty liver disease(7.53, 1.96–28.96), hyperlipidemia(2.15, 1.26–3.67), other lung diseases(6.00, 3.01–11.96), and electrolyte imbalance(10.40, 3.00–26.10)were independently linked to increased odds of being severely ill. T2 DM(6.07, 2.89–12.75), CVD(8.47,6.03–11.89), and electrolyte imbalance(19.44, 11.47–32.96) were also strong predictors of unfavorable outcomes. Women with comorbidities were more likely to have severe disease on admission(5.46,3.25–9.19), while men with comorbidities were more likely to have unfavorable treatment outcomes(6.58, 1.46–29.64) within two weeks. Conclusion Besides hypertension, diabetes, and CVD, fatty liver disease, hyperlipidemia, other lung diseases, and electrolyte imbalance were independent risk factors for COVID-19 severity and poor treatment outcome. Women with comorbidities were more likely to have severe disease, while men with comorbidities were more likely to have unfavorable treatment outcomes.MA Yan ZHU Dong Shan CHEN Ren Bo SHI Nan Nan LIU Si Hong FAN Yi Pin WU Gui Hui YANG Pu Ye BAI Jiang Feng CHEN Hong CHEN Li Ying FENG Qiao GUO Tuan Mao HOU Yong HU Gui Fen HU Xiao Mei HU Yun Hong HUANG Jin HUANG Qiu Hua HUANG Shao Zhen JI Liang JIN Hai Hao LEI Xiao LI Chun Yan LI Min Qing LI Qun Tang LI Xian Yong LIU Hong De LIU Jin Ping LIU Zhang MA Yu Ting MAO Ya MO Liu Fen NA Hui WANG Jing Wei SONG Fang Li SUN Sheng WANG Dong Ting WANG Ming Xuan WANG Xiao Yan WANG Yin Zhen WANG Yu Dong WU Wei WU Lan Ping XIAO Yan Hua XIE Hai Jun XU Hong Ming XU Shou Fang XUE Rui Xia YANG Chun YANG Kai Jun YUAN Sheng Li ZHANG Gong Qi ZHANG Jin Bo ZHANG Lin Song ZHAO Shu Sen ZHAO Wan Ying ZHENG Kai ZHOU Ying Chun ZHU Jun Teng ZHU Tian Qing ZHANG Hua Min WANG Yan Ping WANG Yong Yan 2020Biomedical and Environmental Sciences2020,33,12:8
Behavioral Abnormality along with NMDAR-related CREB Suppression in Rat Hippocampus after Shortwave Exposure显示文摘Objective To estimate the detrimental effects of shortwave exposure on rat hippocampal structure and function and explore the underlying mechanisms. Methods One hundred Wistar rats were randomly divided into four groups(25 rats per group) and exposed to 27 MHz continuous shortwave at a power density of 5, 10, or 30 m W/cm^2 for 6 min once only or underwent sham exposure for the control. The spatial learning and memory, electroencephalogram(EEG), hippocampal structure and Nissl bodies were analysed. Furthermore, the expressions of N-methyl-D-aspartate receptor(NMDAR) subunits(NR1, NR2 A, and NR2 B), c AMP responsive element-binding protein(CREB) and phosphorylated CREB(p-CREB) in hippocampal tissue were analysed on 1, 7, and 14 days after exposure. Results The rats in the 10 and 30 m W/cm^2 groups had poor learning and memory, disrupted EEG oscillations, and injured hippocampal structures, including hippocampal neurons degeneration, mitochondria cavitation and blood capillaries swelling. The Nissl body content was also reduced in the exposure groups. Moreover, the hippocampal tissue in the 30 m W/cm^2 group had increased expressions of NR2 A and NR2 B and decreased levels of CREB and p-CREB. Conclusion Shortwave exposure(27 MHz, with an average power density of 10 and 30 m W/cm^2) impaired rats' spatial learning and memory and caused a series of dose-dependent pathophysiological changes. Moreover, NMDAR-related CREB pathway suppression might be involved in shortwave-induced structural and functional impairments in the rat hippocampus.YU Chao BAI Yan Xin XU Xin Ping GAO Ya Bing HAO Yan Hui WANG Hui TAN Sheng Zhi LI Wen Chao ZHANG Jing YAO Bin Wei DONG Ji ZHAO Li PENG Rui Yun 2019Biomedical and Environmental Sciences2019,32,3:7
Magnified and enhanced computed virtual chromoendoscopy in gastric neoplasia: A feasibility study显示文摘AIM: To evaluate the feasibility of a new computed virtual chromoendoscopy (CVC) device (M i-scan) in the diagnosis of gastric neoplasia. METHODS: Patients with superficial lesions no larger than 1.0 cm found during high definition endoscopy were included. Those with advanced or obviously protruded or depressed lesions, lesions larger than 1.0 cm and/or lesions which were not amenable to observation by zoom function were excluded. The endoscopist was required to give the real-time descriptions of surface pit patterns of the lesions, based on surface pattern classification of enhanced magnification endoscopy. According to previous reports, types Ⅰ-Ⅲ represent nonneoplastic lesions, and types Ⅳ-Ⅴ represent neoplastic lesions. Diagnosis with M i-scan and biopsy was performed before histopathological diagnosis. Magnified images of gastric lesions with and without enhancement were collected for further analysis. The diagnostic yield of real-time M i-scan and effects on magnification image quality by tone enhancement (TE), surface enhancement (SE) and color enhancement (CE) were calculated. The selected images were sent to another endoscopist. The endoscopist rated the image quality of each lesion at 3 levels. Ratings of image quality were based on visualization of pit pattern, vessel and demarcation line. RESULTS: One hundred and eighty-three patients were recruited. Five patients were excluded for advanced gastric lesions, 1 patient was excluded for poor preparation and 2 patients were excluded for superficial lesions larger than 1.0 cm; 132 patients were excluded for no lesions found by high definition endoscopy. In the end, 43 patients with 43 lesions were included. Histopathology revealed 10 inflammation, 14 atrophy, 10 metaplasia, 1 low grade dysplasia (LGD), 5 high grade dysplasia (HGD) and 3 cancers. For 7 lesions classified into type Ⅰ, histopathology revealed 6 atrophy and 1 metaplasia; for 10 lesions classified into type Ⅱ, histopathology revealed 2 inflammation, 7 atrophy and 1 metaplasia; for 10 lesions classified into type Ⅲ, histopathology revealed 1 inflammation, 8 metaplasia and 1 LGD; for 9 lesions classified into type Ⅳ, histopathology revealed 4 inflammation, 1 atrophy and 4 HGD; for 7 lesions classified into type Ⅴ, histopathology revealed 3 inflammation, 1 HGD and 3 cancers. A total of 172 still images, including 43 images by white light (MWL) and 129 images by M i-scan (43 with TE, 43 with SE and 43 with CE), were selected and sent to the endoscopist who did the analysis. General image quality of M i-scan with TE and SE was significantly better than that of MWL (TE, 4.55 ± 1.07; SE, 4.30 ± 1.02; MWL, 3.25 ± 0.99; P < 0.001). Visualization of pit pattern was significantly improved by M i-scan with SE (1.93 ± 0.25 vs 1.50 ± 0.50, P < 0.001). Microvessel visualization was significantly improved by M i-scan with TE (1.23 ± 0.78 vs 0.76 ± 0.73, P < 0.001). Demarcation line visualization was improved by M i-scan with both TE and SE (TE, 1.75 ± 0.52; SE, 1.56 ± 0.59; MWL, 0.98 ± 0.44; P < 0.001). M i-scan with CE did not show any significant improvements of image quality in general or in the 3 key parameters. Although M i-scan with TE and SE slightly increased the diagnostic yield of MWL, there was no significant difference (P > 0.1). CONCLUSION: Although digital enhancement improves the image quality of magnification endoscopy, its value in improving the diagnostic yield seems to be limited.Chang-Qing Li Ya Li Xiu-Li Zuo Rui Ji Zhen Li Xiao-Meng Gu Tao Yu Qing-Qing Qi Cheng-Jun Zhou Yan-Qing Li 2013World Journal of Gastroenterology2013,19,26:5
Genome-wide identification and analysis of AP2/ERF transcription factors related to camptothecin biosynthesis in Camptotheca acuminata显示文摘Camptotheca acuminata produces camptothecin(CPT),a monoterpene indole alkaloid(MIA)that is widely used in the treatment of lung, colorectal, cervical, and ovarian cancers. Its biosynthesis pathway has attracted significant attention, but the regulation of CPT biosynthesis by the APETALA2/ethylene-responsive factor(AP2/ERF)transcription factors(TFs)remains unclear. In this study,a systematic analysis of the AP2/ERF TFs family in C.acuminata was performed, including phylogeny, gene structure, conserved motifs, and gene expression profiles in different tissues and organs(immature bark, cotyledons, young flower, immature fruit, mature fruit, mature leaf, roots, upper stem, and lower stem)of C.acuminata.A total of 198 AP2/ERF genes were identified and divided into five relatively conserved subfamilies, including AP2(26 genes),DREB(61 genes),ERF(92 genes),RAV(18 genes),and Soloist(one gene). The combination of gene expression patterns in different C.acuminata tissues and organs, the phylogenetic tree, the co-expression analysis with biosynthetic genes, and the analysis of promoter sequences of key enzymes genes involved in CPT biosynthesis pathways revealed that eight AP2/ERF TFs in C.acuminata might be involved in CPT synthesis regulation, which exhibit relatively high expression levels in the upper stem or immature bark.Among these, four genes(Cac AP2/ERF123,Cac AP2/ERF125,Cac AP2/ERF126,and Cac AP2/ERF127)belong to the ERF–B2 subgroup;two genes(Cac AP2/ERF149 and Cac AP2/ERF152)belong to the ERF–B3 subgroup;and two more genes(Cac AP2/ERF095 and Cac AP2/ERF096)belong to the DREB–A6 subgroup. These results provide a foundation for future functional characterization of the AP2/ERF genes to enhance the biosynthesis of CPT compounds of C.acuminata.HU Ya-Ting XU Zhi-Chao TIAN Ya GAO Ran-Ran JI Ai-Jia PU Xiang-Dong WANG Yu LIU Xia SONG Jing-Yuan 2020Chinese Journal of Natural Medicines2020,18,8:5
Estimating biological reference points for Largehead hairtail(Trichiurus lepturus) fishery in the Yellow Sea and Bohai Sea显示文摘It is important to find a reliable method to estimate maximum sustainable yield(MSY)or total allowable catch(TAC)for fishery management, especially when the data availability is limited which is a case in China.A recently developed method(CMSY)is a data-poor method, which requires only catch data, resilience and exploitation history at the first and final years of the catch data.CMSY was used in this study to estimate the biological reference points for Largehead hairtail(Trichiurus lepturus,Temminck and Schlegel)in the Yellow Sea and Bohai Sea,based on the fishery data from China Fishery Statistical Year Books during 1986 to 2012.Additionally,Bayesian state-space Schaefer surplus production model(BSM)and the classical surplus production models(Schaefer and Fox)performed by software CEDA and ASPIC,were also projected in this study to compare with the performance of CMSY.The estimated MSYs from all models are about 19.7×104–27.0×104 t,while CMSY and BSM yielded more reasonable population parameter estimates(the intrinsic population growth rate and the carrying capacity). The biological reference points of B/BMSY smaller than 1.0,while F/FMSY higher than 1.0 revealed an over-exploitation of the fishery, indicating that more conservative management strategies are required for Largehead hairtail fishery.Yupeng Ji Qun Liu Baochao Liao Qingqing Zhang Ya’nan Han 2019Acta Oceanologica Sinica2019,38,10:5
Microwave-induced Apoptosis and Cytotoxicity of NK Cells through ERK1/2 Signaling显示文摘Objective To investigate microwave-induced morphological and functional injury of natural killer(NK) cells and uncover their mechanisms. Methods NK-92 cells were exposed to 10, 30, and 50 m W/cm^2 microwaves for 5 min. Ultrastructural changes, cellular apoptosis and cell cycle regulation were detected at 1 h and 24 h after exposure. Cytotoxic activity was assayed at 1 h after exposure, while perforin and NKG2 D expression were detected at 1 h, 6 h, and 12 h after exposure. To clarify the mechanisms, phosphorylated ERK(p-ERK) was detected at 1 h after exposure. Moreover, microwave-induced cellular apoptosis and cell cycle regulation were analyzed after blockade of ERK signaling by using U0126. Results Microwave-induced morphological and ultrastructural injury, dose-dependent apoptosis(P < 0.001) and cell cycle arrest(P < 0.001) were detected at 1 h after microwave exposure. Moreover, significant apoptosis was still detected at 24 h after 50 m W/cm^2 microwave exposure(P < 0.01). In the 30 m W/cm^2 microwave exposure model, microwaves impaired the cytotoxic activity of NK-92 cells at 1 h and down regulated perforin protein both at 1 h and 6 h after exposure(P < 0.05). Furthermore, p-ERK was down regulated at 1 h after exposure(P < 0.05), while ERK blockade significantly promoted microwave-induced apoptosis(P < 0.05) and downregulation of perforin(P < 0.01). Conclusion Microwave dose-dependently induced morphological and functional injury in NK-92 cells, possibly through ERK-mediated regulation of apoptosis and perforin expression.ZHAO Li LI Jing HAO Yan Hui GAO Ya Bing WANG Shui Ming ZHANG Jing DONG Ji ZHOU Hong Mei LIU Shu Chen PENG Rui Yun 2017Biomedical and Environmental Sciences2017,30,5:5
Hexagonal boron nitride nanosheet for effective ambient N2 fixation to NH3显示文摘Industrial production of NH3 from N2 and H2 significantly relies on Haber-Bosch process, which suffers from high energy consume and CO2 emission. As a sustainable and environmentally-benign alternative process, electrochemical artificial N2 fixation at ambient conditions, however,is highly required efficient electrocatalysts. In this study,we demonstrate that hexagonal boron nitride nanosheet (h-BNNS) is able to electrochemically catalyze N2 to NH3.In acidic solution,h-BNNS catalyst attains a high NH3 formation rate of 22.4 μg·h-1·mg-1cat.and a high Faradic efficiency of 4.7% at-0.75 V vs.reversible hydrogen electrode, with excellent stability and durability.Density functional theory calculations reveal that unsaturated boron at the edge site can activate inert N2 molecule and significantly reduce the energy barrier for NH3 fonmation.Ya Zhang Huitong Du Yongjun Ma Lei Ji Haoran Guo Ziqi Tian Hongyu Chen Hong Huang Guanwei Cui Abdullah M. 16Preparation and evaluation of a new releasable PEGylated tumor necrosis factor-α(TNF-α) conjugate for therapeutic application显示文摘To design a releasable PEGylated TNF-α(rPEG-TNF-α ), a cathepsin B-sensitive dipeptide (Val-Cit moiety) was inserted into conventional PEG-modified TNF- (PEG-TNF- ), facilitating its clinical use for anti-tumor therapy. Comparative pharmaco- kinetic and pharmacodynamic studies showed that the half-lives of both PEGylated forms of TNF-α were ~60-fold greater than that of unmodified TNF-α . In addition, the in vitro bioactivity of rPEG-TNF-α was greater than that of PEG-TNF-α with the same degree of PEG modification. Release of TNF-α from rPEG-TNF-α in vitro was dependent on the presence of cathepsin B and was inhibited by a cathepsin B inhibitor. Despite the potent cytotoxicity of unmodified TNF-α against normal cells, its PEGylated forms at higher TNF-α concentrations showed low cytotoxic activity against these cells. In contrast, both forms of PEGylated TNF-α showed potent cytotoxic activity against the B16 and L929 cell lines, with rPEG-TNF-α being 5- and 9- fold more potent, respectively, than PEG-TNF-α . Moreover, rPEG-TNF-α was a more potent in vivo antitumor agent than PEG-TNF-α .DAI ChuanYun FU Ya CHEN ShaoCheng LI Biao YAO Bo LIU WanHong ZHU LiQing CHEN Nan CHEN Ji ZHANG Qiang 2013Science China(Life Sciences)2013,56,1:4
Effects of Electroacupuncture Intervention on Expression of Cyclooxygenase 2 and Microglia in Spinal Cord in Rat Model of Neuropathic Pain显示文摘Objective: To investigate the effect of electroacupuncture(EA) treatment on the expression of cyclooxygenase(COX) 2 and microglia in spinal cord by using rat model of neuropathic pain, and to probe into the relationship between COX 2 and microglia. Methods: The rats were randomly divided into 6 groups, including normal control group, model group, sham group, EA 1 group(distant acupoints + local acupoints), EA 2 group(local acupoints), and EA 3 group(distant acupoints). Thermal withdrawal latencies were evaluated at 1 day preoperatively and 3, 5 and 7 days postoperatively. At 7 days postoperatively, the spinal COX 2 m RNA was detected by reverse-transcription polymerase chain reaction. Double immunofluorescent staining technology was applied to screen and verify the relationship between altered COX 2 and microglia. Results: Compared with the model group, thermal withdrawal latencies increased after EA treatment(P<0.01). The expressions of COX 2 m RNA were up-regulated in spinal cord of rat on day 7 after surgery(P<0.05). Compared with the model group, EA stimulation(EA 1 and EA 2 groups) reversed the up-regulation of COX 2 m RNA expression(P<0.05). EA 1 and EA 2 groups might have better treatment effect compared with the EA 3 group. Fluorescent images displayed COX 2 and microglia expressed at common areas. Conclusions: EA was effective in analgesic and anti-inflammatory. EA has decreased the expression of spinal COX 2 m RNA in the trend of the therapeutic effect of 'distant acupoints + local acupoints', and 'local acupoints' intervention may be superior to that of 'distant acupoints' intervention. Microglia may be related to the formation of COX 2.JI Li-li GUO Meng-wei REN Xiu-jun GE Dong-yu LI Gen-mao TU Ya 2017Chinese Journal of Integrative Medicine2017,23,10:4
Preparation and performance of lipophilic α-zirconium phosphate with high thermal stability and its application in thermal-plastic polymers显示文摘To prepare lipophilic α-zirconium phosphate with high grafting ratio and thermal stability(OZrP-HT) and explore its potential application in thermal-plastic polymers,a novel method was developed by surface lipophilicity enhancement strategy. The commercial a-zirconium phosphate(a-ZrP) was pre-intercalated by n-propylamine(PA) and grafted by silane coupling agents. Then the pre-intercalated PA was removed by heattreatment, and the obtained OZrP-HT was utilized to fabricate the phosphorous-containing polyester(P-co-PET)/OZrP-HT nanocomposites by melt-blending method. The prepared OZrP-HT and P-co-PET/OZrP-HT nanocomposites were characterized by Wide Angle X-ray Diffraction(WAXD),Fourier Transform Infrared Spectroscopy(FTIR),Thermogravimetric Analysis(TGA),Transmission Electron Microscope(TEM),etc. The results show that OZrP-HT with high grafting ratio(13.78 wt%) and thermal stability(T_(onset)= 368℃) was successfully prepared via this novel method and was uniformly intercalated by P-co-PET molecular chains.OZrP-HT had no significant effect on the fiber processability of P-co-PET polymer, and flame retardant properties of(P-co-PET)/OZrP-HT nanocomposites were improved. This method may be suitable for organic modification of general inorganic layered compounds and could extend the potential applications in thermo-plastic polymers.Ya Du Feng Deng Xiaoze Jiang Huquan Ji Dan Yu Wei Wang Bin Sun Meifang Zhu 2015Progress in Natural Science:Materials International2015,25,5:3
Determination of the metastable zone and induction time of thiourea for cooling crystallization显示文摘The solubility, metastable zone width(MSZW),and induction time of thiourea for cooling crystallization were experimentally determined in the temperature range of 283-323 K.The solubility data could be well described by the Apelblat equation model as lnx=-99.55+1071.66/T+16.27 lnT.The determinations of the effects of various stirring and cooling rates indicated that the MSZW increased with increasing and decreasing cooling and stirring rates, respectively.Furthermore, the induction times at various temperatures and supersaturation ratios were also measured. The results indicated that homogeneous nucleation could occur at high supersaturation, whereas heterogeneous nucleation was more likely to occur at low supersaturation.Based on the classical nucleation theory and induction period data, the calculated solid-liquid interfacial tensions of thiourea in deionized water at 302.46 and 312.58 K were 2.86 and 2.94 mJ·m^(-2),respectively.Tong Zhou Chunzhao Tu Ya Sun Linan Ji Chuangxian Bian Xiaohua Lu Changsong Wang 2021Chinese Journal of Chemical Engineering2021,34,3:3
