1Insulin improves cardiac myocytes contractile function recovery in simulated ischemia-reperfusion:Key role of Akt显示文摘The present study examined cardiac myocytecontractile and Ca2+ transient responses to insulin duringsimulated ischemia/reperfusion (I/R) and furtherinvestigated the role of protein kinase B (Akt) in the insulin-induced inotropic effect. Ventricular myocytes wereenzymatically isolated from adult Sprague-Dawley rats and perfused with Tyrode solution while electrically field-stimulated. Simulated I/R was induced by perfusing the cells with chemical anoxic solution including sodiumcyanide-sodium lactate for 15 min followed by reperfusionwith normal oxygenated Tyrode solution with or withoutinsulin. It is found that insulin only at concentration as high as 10 IU/L could increase cell shortening (16±5%, P < 0.05) in normal myocytes, whereas it concentration-dependently (0.01—10 IU/L) increased the contraction,the velocity ofshortening/releng- thening and Ca2+ transient in I/Rmyocytes. In addition, insulin treatment (1 IU/L) increasedAkt phosphorylation of I/R cardiomyocytes by 2.4-foldcompared with that of the control (P < 0.01). Mostimportantly, pretreatment with LY 294002, a specificinhibitor of phosphatidylinositol 3′-kinase (PI3-kinase), significantly inhibited both Akt phosphorylation and thepositive inotropic response to insulin in the I/Rcardiomyocytes. These results suggest that insulin exertsdirect positive inotropic effect by increasing Ca2+ transient of cardiomyocytes, which is enhanced in the pathologicalcondition of I/R. 3Effects of reclaimed water irrigation and nitrogen fertilization on the chemical properties and microbial community of soil显示文摘The ecological effect of reclaimed water irrigation and fertilizer application on the soil environment is receiving more attention.Soil microbial activity and nitrogen(N)levels are important indicators of the effect of reclaimed water irrigation on environment.This study evaluated soil physicochemical properties and microbial community structure in soils irrigated with reclaimed water and receiving varied amounts of N fertilizer.The results indicated that the reclaimed water irrigation increased soil electrical conductivity(EC)and soil water content(SWC).The N treatment has highly significant effect on the ACE,Chao,Shannon(H)and Coverage indices.Based on a 16S ribosomal RNA(16S rRNA)sequence analysis,the Proteobacteria,Gemmatimonadetes and Bacteroidetes were more abundant in soil irrigated with reclaimed water than in soil irrigated with clean water.Stronger clustering of microbial communities using either clean or reclaimed water for irrigation indicated that the type of irrigation water may have a greater influence on the structure of soil microbial community than N fertilizer treatment.Based on a canonical correspondence analysis(CCA)between the species of soil microbes and the chemical properties of the soil,which indicated that nitrate N(NO_3~–-N)and total phosphorus(TP)had significant impact on abundance of Verrucomicrobia and Gemmatimonadetes,meanwhile the p H and organic matter(OM)had impact on abundance of Firmicutes and Actinobacteria significantly.It was beneficial to the improvement of soil bacterial activity and fertility under 120 mg kg^(–1) N with reclaimed water irrigation.GUO Wei Mathias N Andersen QI Xue-bin LI Ping LI Zhong-yang FAN Xiang-yang ZHOU Yuan 2017Journal of Integrative Agriculture2017,16,3:7

5Color 3D Reverse Engineering显示文摘This paper presents a principle and a method of col or 3D laser scanning measurement. Based on the fundamental monochrome 3D measureme nt study, color information capture, color texture mapping, coordinate computati on and other techniques are performed to achieve color 3D measurement. The syste m is designed and composed of a line laser light emitter, one color CCD camera, a motor-driven rotary filter, a circuit card and a computer. Two steps in captu ring object’s images in the measurement process: First, the color CCD camera ta kes the color picture of the object to be used for texture mapping in post proce ssing. Secondly, the monochrome data of the object is taken. The laser light emi tter is started; the light plane and the surface of the object intersect to form an undulate line, which forms an image on the CCD sensor. After the optic- electronics transformation, the electronic signals are captured and send to the computer. A new color 3D measurement model was derived, and a rapid calibra ting method to measure the system parameters was proposed--optical plane equat ion calibrating method. A calibrating drone was designed and built. 3D color sca nning system not only provides an object’s individual point information in the 3D coordinate, in the mean time, it provides the color information of each indiv idual point. This paper also advances a multi-frame auto-merging method, i.e. several frames of color 3D digital images measured are merged quickly according to their curvature characteristics and RGB information. Initial matching and fin e tune of the registration are completely performed by the computer; initial mat ching is via user interface in helping computer to find the transformation matri x. The transformation matrix is found according to the geometric characteristics chosen by hand. After the initial transformation matrix is found, fine tune of the registration is preformed to do the optimum adjustment to achieve a complete color textured 3D model.
