| 1 | Recombinant Newcastle disease virus expressing African swine fever virus protein 72 is safe and immunogenic in mice显示文摘African swine fever(ASF) is a lethal hemorrhagic disease that affects wild and domestic swine. The etiological agent of ASF is African swine fever virus(ASFV). Since the first case was described in Kenya in 1921, the disease has spread to many other countries. No commercial vaccines are available to prevent ASF. In this study, we generated a recombinant Newcastle disease virus(r NDV) expressing ASFV protein 72(p72) by reverse genetics and evaluated its humoral and cellular immunogenicity in a mouse model. The recombinant virus, r NDV/p72, replicated well in embryonated chicken eggs and was safe to use in chicks and mice. The p72 gene in r NDV/p72 was stably maintained through ten passages. Mice immunized with r NDV/p72 developed high titers of ASFV p72 specific Ig G antibody, and had higher levels of Ig G1 than IgG2 a. Immunization also elicited T-cell proliferation and secretion of IFN-γ and IL-4. | 3 | Effects of Bacillus subtilis CSL2 on the composition and functional diversity of the faecal microbiota of broiler chickens challenged with Salmonella Gallinarum显示文摘Background: The chicken gastrointestinal tract contains a diverse microbiota whose composition and structure play important roles in gut functionality. In this study, microbial shifts resulting from feed supplementation with Bacillus subtilis CSL2 were evaluated in broilers challenged and unchallenged with Salmonella Gallinarum. To analyse bacterial community composition and functionality, 454 GS-FLX pyrosequencing of 16 S r RNA gene amplicons was performed.Results: The Quantitative Insights into Microbial Ecology(QIIME) pipeline was used to analyse changes in the faecal microbiota over a 24-h period. A total of 718,204 sequences from broiler chickens were recorded and analysed. At the phylum level, Firmicutes, Bacteroidetes, and Proteobacteria were the predominant bacterial taxa. In Salmonellainfected chickens(SC), Bacteroidetes were more highly abundant compared to control(NC) and Bacillus-treated(BT)chickens. At the genus level, in the NC and BT groups, Lactobacil us was present at high abundance, and the abundance of Turicibacter, unclassified Enterobacteriaceae, and Bacteroides increased in SC broilers. Furthermore, taxon-independent analysis showed that the SC and BT groups were compositional y distinct at the end of the 24-h period. Further analysis of functional properties showed that B. subtilis CSL2 administration increased gut-associated energy supply mechanisms(i.e. carbohydrate transport and metabolism) to maintain a stable microbiota and protect gut integrity.Conclusions: This study demonstrated that S. Gallinarum infection and B. subtilis CSL2 supplementation in the diet of broiler chickens influenced the diversity, composition, and functional diversity of the faecal microbiota. | 6 | Influence of(FeO+Ti O_2) abundance on the microwave thermal emissions of lunar regolith显示文摘One of the essential controls on the microwave thermal emissions(MTE) of the lunar regolith is the abundance of Fe O and TiO_2, known as the(Fe O+Ti O_2) abundance(FTA). In this paper, a radiative transfer simulation is employed first to study the change in the brightness temperature(T_B) with FTA under a range of frequencies and surface temperatures. Then, we analyze the influence of FTA on the MTE of the lunar regolith using microwave sounder(CELMS) data from the Chang'E-2 lunar orbiter, Clementine UV-VIS data, and lunar samples recovered from the Apollo and Surveyor projects. We conclude that:(1) FTA strongly influences the MTE of the lunar regolith, but it is not the decisive control, and(2) FTA decreases slightly with depth. | 11 | Bendamustine treatment of Chinese patients with relapsed indolent non-Hodgkin lymphoma: a multicenter, open-label, single-arm, phase 3 study显示文摘Background:Bendamustine was approved in China on May 26th,2019 by the National Medical Product Administration for the treatment of indolent B-cell non-Hodgkin lymphoma(NHL).The current study was the registration trial and the first reported evaluation of the efficacy,safety,and pharmacokinetics of bendamustine in Chinese adult patients with indolent B-cell NHL following relapse after chemotherapy and rituximab treatment.Methods:This was a prospective,multicenter,open-label,single-arm,phase 3 study(NCT01596621;C18083/3076)with a 2-year follow-up period.Eligible patients received bendamustine hydrochloride 120 mg/m2 infused intravenously on days 1 and 2 of each 21-day treatment cycle for at least six planned cycles(and up to eight cycles).The primary endpoint was the overall response rate(ORR);and secondary endpoints were duration of response(DoR),progression-free survival(PFS),safety,and pharmacokinetics.Patients were classified according to their best overall response after initiation of therapy.Proportions of patients in each response category(complete response[CR],partial response[PR],stable disease,or progressive disease)were summarized along with a twosided binomial exact 95%confidence intervals(CIs)for the ORR.Results:A total of 102 patients were enrolled from 20 centers between August 6th,2012,and June 18th,2015.At the time of the primary analysis,the ORR was 73%(95%CI:63%–81%)per Independent Review Committee(IRC)including 19%CR and 54%PR.With the follow-up period,the median DoR was 16.2 months by IRC and 13.4 months by investigator assessment;the median PFS was 18.6 months and 15.3 months,respectively.The most common non-hematologic adverse events(AEs)were gastrointestinal toxicity,pyrexia,and rash.Grade 3/4 neutropenia was reported in 76%of patients.Serious AEs were reported in 29 patients and five patients died during the study.Pharmacokinetic analysis indicated that the characteristics of bendamustine and its metabolites M3 and M4 were generally consistent with those reported for other ethnicities.Conclusion:Bendamustine is an active and effective therapy in Chinese patients with relapsed,indolent B-cell NHL,with a comparable risk/benefit relationship to that reported in North American patients. | Yuan-Kai Shi Xiao-Nan Hong Jian-Liang Yang Wei Xu Hui-Qiang Huang Xiu-Bin Xiao Jun Zhu Dao-Bin Zhou Xiao-Hong Han Jian-Qiu Wu Ming-Zhi Zhang Jie Jin Xiao-Yan Ke Wei Li De-Pei Wu Shen-Miao Yang Xin Du Yong-Qian Jia Ai-Chun Liu Dai-Hong Liu Zhi-Xiang Shen Lian-Sheng Zhang Leonard James Edward Hellriegel | 2021 | Chinese Medical Journal2021，，11： | 2 |
| 12 | ANALYSIS OF FUNGI IN MUSHROOM COMPOST USING PCR AND TGGE OF RIBOSOMAL DNA显示文摘The fungal communities in mushroom compost phase Ⅱ was assessed using a combination of PCR amplification and sequencing of 18S rDNA from fungal isolates and “nested” PCR TGGE analysis on the basis of DNA directly extracted from compost samples. The diversity of cultivated fungi isolated from compost samples was low. A total of 11 isolates were related to only 2 different species. One species, Chaetomium elatum, was identified within 10 isolates, and the other, with high similarity belonged to Penicillium expansum. The fungal flora associated with mushroom compost was then monitored with “nested” PCR TGGE.
