1Methodologic research on TIMP-1,TIMP-2 detection as a new diagnostic index for hepatic fibrosis and its significance显示文摘AIM: To set up a new method to detect tissue inhibitors ofmetalloproteinase1 and -2(TIMP-1 and TIMP-2) in sero ofpatients with hepatic cirrhosis, and to investigate theexpression and location of TIMP-1 and TIMP-2 in liver tissueof patients with hepatic cirrhosis, and the correlationbetween TIMPs in liver and those in sera so as to discusswhether TIMPs can be used ss a diagnosis index of hepaticfibrosisMETHODS: The monoclonal antibodies (McAbs) of TIMP-1and TIMP-2 were used to sensitize erythrocytes, and solid-phase absorption to sensitized erythrocytes (SPASE) wasused to detect TIMP-1 and TIMP-2 in the sera of patients withhepatic cirrhosis. Meanwhile, with the method of in situhybridization and immunohistochemistry, we studied themRNA expression and antigen location of TIMP-1 and TIMP-2in the livers of 40 hepatic cirrhosis patients with pathologicdiagnosis.RESULTS: With SPASE, they were 16.4 % higher in theacute hepatitis group, 33.3 % higher in the chronic hepatitisgroup, and the positive rates were 73.6 % and 61. 2 %respectively in sero of hepatic cirrhosis patients, which wereremarkably higher than those in chronic hepatitis and acutehepatitis group ( P < 0. 001 ). In 40 samples of hepaticcirrhosis tissues, all of them showed positive expression ofTIMP-1 and TIMP-2 mRNA detected withimmunohistochemistry or in situ hybridization (positive ratewas 100 % ). Expression of TIMPs in different degrees couldbe found in liver tissue with cirrhosis. 2SHRIMP zircon U-Pb age and Nd isotopic study on the Nyainqêntanglha Group in Tibet显示文摘The Nyainqêntanglha Group is traditionally viewed as the oldest metamorphic basement in the Lhasa block, but its formation age and tectonic setting remain debate. Zircons extracted from the metamorphic sequence of volcanics and intrusions of the Nyainqêntanglha Group, 10 km west of Nam Co in northern Lhasa block, have been investigated by cathodolu- minescence (CL), backscattered (BSE) and dated by ion microprobe (SHRIMP). We conclude that the U-Pb age of 787±9 Ma of zircons from the trondhjemite imposes a constraint on maxi- mum protolith age, and minimum formation age of the Nyainqêntanglha Group is constrained by U-Pb age of 748±8 Ma of zircons from the granite. The formation age of the Nyainqêntanglha Group is consistent with sedimentary age of Greater Himalayan rocks, showing that they devel- oped coevally in an arc-basin tectonic setting of Neoproterozoic active continental margin along the northern margin of the India shield. The inherited zircons from the tholetiite and granite give older 207Pb/206Pb ages from 947 to 1766 Ma. The positive εNd( t ) value indicates that the mafic rocks were derived from the depleted mantle, but contaminated by the older continental crustal material. 3Early J_2 basalts in SE China:Incipience of large-scale late Mesozoic magmatism显示文摘Magmatism in SE China was dormant during 204―180 Ma, but was reactivated in 180― 170 Ma (early J2), and then became more and more intensive towards the end of early Cretaceous. The small-scale early J2 magmatism is the incipience to long-term and large-scale magmatism in this region. A near east-west (EW) trend volcanic belt was distributed across south Hunan, south Jiangxi and southwest Fujian was formed during early J2 time. Along this belt from the inland toward the coast, the lithology of basalts changes from alkali into tholeiite, and the amount of erupted volcanic rocks and the proportions of rhyolites coexisting with the basalts increase. On the basis of geochemical char- acteristics of these basalts, we infer that the melting degree of source rocks and the extent of frac- tional crystallization and crustal contamination all increased whereas the depth of mantle source de- creased from the inland to the coast, which led to the variations of geological characteristics of the volcanic belt. In early J2, the western spreading Pacific plate began to subduct underneath SE China continental block, reactivating near EW trend deep fault that was originally formed during the Indos- inian event. 5Structure and function of aggrecan显示文摘Aggrecan is the major proteoglycan in the articular cartilage. This molecule is important in the proper functioning of articular cartilage because it provides a hydrated gel structure (via its interaction with hyaluronan and link protein) that endows the cartilage with load-bearing properties. It is also crucial in chondroskeletal morphogenesis during development. Aggrecan is a multimodular molecule expressed by chondrocytes. Its core protein is composed of three globular domains (G1, G2, and G3) and a large extended region (CS) between G2 and G3 for glycosaminoglycan chain attachment. G1 comprises the amino terminus of the core protein. This domain has the same structural motif as link protein. Functionally, the G1 domain interacts with hyaluronan acid and link protein, forming stable ternary complexes in the extracellular matrix. G2 is homologous to the tandem repeats of G1 and of link protein and is involved in product processing. G3 makes up the carboxyl terminus of the core protein. It enhances glycosaminoglycan modification and product secretion. 7The characteristics of atmospheric CO_2 concentration variation of four national background stations in China显示文摘Atmospheric carbon dioxide is an important kind of greenhouse gas which influences global temperature. Its concentration variation could indicate the distribution of human and natural activities in various regions. Through the non-dispersive infrared method, flask sampling of atmospheric CO2 concen- tration was measured weekly at four national background stations including Waliguan, Shangdianzi, Lin’an, and Longfengshan. Based on the data collected from September 2006 to August 2007, along with the Waliguan station’s experience on in situ observational data processing, the selection methods for sampling data through the atmospheric background CO2 concentration analysis were preliminarily discussed. On the basis of this result, the variation features of the four typical regions’ atmospheric background CO2 concentration was analyzed for the first time. The results show that the atmospheric CO2 concentration at Waliguan, Shangdianzi, Lin’an, and Longfengshan is 383.5, 385.9, 387.8, and 384.3 ppm, respectively. During the research period, CO2 concentration at the Waliguan station changed slightly. However, the CO2 concentration changed sharply at the Shangdianzi and the Lin’an stations due to the great influence of human activities in the Jingjinji and the Changjiang Delta economic zones, and changed regularly with seasons at Longfengshan station under dual influences of human activities and plant photosynthesis. 10Investigation of changes in properties of water under the action of a magnetic field显示文摘The properties of water and their changes under the action of a magnetic field were gathered by the spectrum techniques of infrared, Raman, visible, ultraviolet and X-ray lights, which may give an insight into molecular and atomic structures of water. It was found that some properties of water were changed, and a lot of new and strange phenomena were discovered after magnetization. Magnetized water really has magnetism, which has been verified by a peak shift of X-ray diffraction of magnetized water + Fe3O4 hybrid relative to that of pure water + Fe3O4 hybrid, that is a saturation and memory effect. The properties of infrared and ultraviolet absorp-tions, Raman scattering and X-ray diffraction of magnetized water were greatly changed relative to those of pure water; their strengths of peaks were all increased, the frequencies of some peaks did also shift, and some new peaks, for example, at 5198, 8050 and 9340 cm?1, occurred at 25℃ after water was magnetized. In the meanwhile, the magnetized effects of water are related to the magnetized time, the intensity of an externally applied magnetic field, and the temperature of water, but they are not a linear relationship. The study also showed a lot of new and unusual properties of magnetized water, for example, the six peaks in 3000-3800 cm?1 in infrared absorption, the exponential increase of ultraviolet absorption of wave with the decreasing wavelength of light of 200-300 nm, the frequency-shifts of peaks, a strange irreversible effect in the increasing and decreasing processes, as well as a stronger peak of absorption occurring at 50℃, 70℃ and 80℃, the existence of many models of motion from 85℃ to 95℃ in 8000-10000 cm?1, and so on. These results show that the molecular structure of water is very complicated, which needs further study. Furthermore, the macroscopic feature of mechanics, for instance, surface tension force of magnetized water, was also measured. Experiments dis-covered that the size in contact angles of magnetized water on the surface of hy-drophobic materials decreases, thus the surface tension force of magnetized water decreases relative to that of pure water. It is seen from the above results that the clustering structure of hydrogen-bonded chains and polarization effects of water molecules are enhanced after magnetization. 11Presence of Antibodies to Heat Stress Proteins and its Possible Significance in Workers Exposed to High Temperature and Carbon Monoxide显示文摘Antibodies to the ubiquitous group of stress proteins known as heat shock proteins (Hsps) have been found to be associated with a number of diseases in humans. Hsps are known to be induced by certain xenobiotics, some of which are common in the working environment. The biological significance of the presence of such autoantibodies is presently unclear. In the present study, we used immunoblotting to investigate the presence of antibodies against the different stress proteins, Hsp27, Hsp60, Hsp71, Hse (heat shock cognate ) 73 and Hsp89a and D in groups of workers exposed to high temperature or carbon monoxide. These data were related to a detailed clinical evaluation and to various laboratory measurements including electrocardiogram (ECG), B echogram, white blood cell counts and typing, the activity of alanine aminotransferase (ALT), acid phosphatase (ACP) and alkaline phosphatase (ALP) and lymphocyte DNA damage. Antibodies to Hsp27 and Hsp71 were found more frequently in the high temperature and carbon monoxide-exposed groups than in controls (P (0.05 ). The carbon monoxide-exposed group showed the highest incidence of anti-Hsp antibodies. Anti-Hsp60 antibodies were only detected in workers exposed to high temperature or carbon monoxide. The percentage of workers with abnormal ECG, B echogram changes and displaying hepatitis B antigen (HBsAg ) was higher in the carbon monoxide group than in the control group (P<0.05 ).There was a significant inerease in the activity of ALT in the high temperature and carbon monoxide groups and in the activities of ACP and ALP in the carbon monoxide group (P<0.05 ). The extent of DNA damage measured in lymphoeytes was higher in workers from the high temperature and carbon monoxide-cxposed groups. We suggest that the increased frequeney of antibodies to Hsps is the result of these damages, of the release of denatured Hsps and of a decrease in the phagocytic ability of macrophages in these workers. 12Ensemble projection of 1―3℃ warming in China显示文摘Studies on the influences of climate change on biogeochemical cycles and on the key vulnerabilities and the risk from climate change suggest that annual surface temperature rise of 1℃, 2℃ and 3℃ above the present level would lead to changes in extreme weather and climate events, food production, fresh water resources, biodiversity, human mortality, etc. Here two sets of simulations as performed with seventeen atmosphere-ocean general circulation models (AOGCMs) for the Fourth Assessment Report of the Intergovernmental Panel on Climate Change (IPCC AR4), i.e. the model outputs from the 20th Century Climate in Coupled Models (20C3M) and from the Special Report on Emissions Scenarios (SRES) emission scenarios B1, A1B and A2, are used to analyze spatial and temporal characteristics of the above values in China over the 21st century. The results indicate that the rate of warming varies from region to region. The above values are reached much later (earlier) when emission amount is lower (higher), and spread of the time when the lower (higher) value is exceeded is narrower (wider) among the three scenarios. 13Efficacy of intramuscular matrine in the treatment of chronic hepatitis B显示文摘BACKGROUND: Hepatitis B virus (HBV) infection, a glo- bal public health problem, is the leading cause of cirrhosis and hepatocellular carcinoma (HCC) worldwide. There are more than 350 million HBV carriers in the world and up to one million die annually due to hepatitis B associated liv- er disease. So far no optimal treatment is available for pa- tients with chronic hepatitis B. In the paper we investigated the efficacy of intramuscular matrine in the treatment of chronic hepatitis B. METHODS: One hundred and twenty patients with chronic hepatitis B were randomly divided into matrine treatment group (n =60) and control group (n =60). The patients of the matrine group were given intramuscularly with matrine (an alkaloid extracted from a traditional Chinese herb Radix Sophorae Flavescentis by Guangzhou Ming Xing Pharmaceu cal Factory, Guangzhou, China) of 100 mg daily for 90 days in addition to conventional liver-protective drugs in- cluding glucurone, inosine, compound vitamin B and caryophyllin. The control group received conventional liv- er-protective drugs alone. Clinical manifestations and labo- ratory parameters including liver biochemistry and serum hepatitis B virus markers were monitored before and after treatment in the two groups. RESULTS: Significant differences were seen between the two groups in terms of improvement of clinical symptoms and signs, recovery of liver functions, and serum conver- sion from hepatitis Be antigen to HBe antibody and from positive to negative serum HBV DNA (P <0.05-0.01). The result of the matrine group was more marked than that of the control group. Serious side-effects were not observed except mild pain at the site of injection of matrine in a few patients. 14Prevention and treatment of hepatic artery thrombosis after liver transplantation显示文摘BACKGROUND: Hepatic artery thrombosis (HAT) which is a serious complication after orthotopic liver transplanta- tion (OLT) remains a significant cause of graft loss. The purpose of this study was to sum up our experiences in the prevention, diagnosis and management of HAT after liver transplantation. METHODS: From April 1993 to September 2003, a total of 198 patients underwent OLT at our hospital. The hepatic artery was anastomosed using 7/0 prolane with running continuous suture in 96 patients (group 1) and with inter- rupted suture in 102 (group 2). Ultrasonography was per- formed every day in two weeks after operation and selec- tively afterwards. RESULTS: HAT occurred in 6 patients (6.3%, 6/96) of group 1, and in 1 (1%, 1/102) of group 2 (x2=4.027, P= 0.045). Six patients received emergency thrombectomy, and 1 conservative therapy but died from tumor recurrence eventually. Biliary complication developed in 3 patients af- ter thrombectomy of whom 2 died of liver failure and one waited for retransplantation. In the other 3 patients after thrombectomy, 1 died of renal failure, and 2 survived. The mortality of patients with HAT was 57.1% (4/7). CONCLUSIONS: The technique of hepatic arterial anasto- mosis is the key factor for the prevention of HAT. Routine ultrasonography is very important in early detection of HAT after OLT. 15Plasmacytoid Dendritic Cells Act as the Most Competent Cell Type in Linking Antiviral Innate and Adaptive Immune Responses显示文摘Appropriate in vivo control of plasmacytoid dendritic cell (pDC) recruitment and activation is a fundamental requirement for defense against viral infection. During this process, a pivotal event that influences the outcome of viral infection is the production of high levels of type I interferon by pDCs. In particular, recent research findings showed that pDCs not only shape the nature of innate resistance, but are also responsible for the successful transition from innate to adaptive immunity for viral resistance. In addition, pDCs can differentiate into antigen presenting cells that may regulate tolerance to a given pathogen. Importantly, in a series of recent clinical studies,pDCs appeared to be defective in number and function in conditions of chronic viral diseases such as infected with HIV-1, HBV or HCV. pDC-associated clinical antiviral therapy is also emerging. 16Chemopreventive Effects of Black Tea Polyphenols in Mouse Skin Model of Carcinogenesis显示文摘In the present investigations, the antitumorigenic effect of black tea polyphenols (BTP) in twcrstage mouse skin model of carcinogenesis was studied. The animals were initiated with a single 'subcarcinogenic' topical dose (52 μg/200 μl acetone ) of 7, 12-dimethylbenzanthracene (DMBA). To evaluate the anti-tumour initiating activity, BTP was topically applied twice a week for three weeks prior to DMBA application, followed by topical treatment with 12-o-tetradecanoyl phorbol-13-acetate (TPA) (5 μg/200 μl acetone, 2x/wk. ) as promoter. For evaluation of antitumor promoting activity, BTP was applied prior to each treatment of TPA. BTP application showed marked inhibitory effect as antitumour initiator as well as antitumour promoter in mouse skin medel of two-stage carcinogenesis. 17Field measurement and analysis of turbulence coherence for Typhoon Nuri at Macao Friendship Bridge显示文摘For the purpose of investigating the turbulent and spatial coherent characteristics of strong wind during typhoon landing period,two 3-dimensional ultrasonic anemometer stations were set up 30 m horizontally apart on the Macao Friendship Bridge to caputure the turbulent wind velocities of Typhoon Nuri. Based on the reliable and representative field measured data,the mean wind speed and direction,turbulence intensity,turbulence integral scale,turbulence power spectra,spatial correlation coefficient and coherence function were statistically evaluated. The field measurement analysis have presented the following results: 1) Two anemometer stations provided consistent results. The mean wind speed variation in time domain presented typical M-shape curves. The strong wind (10-minute mean wind speed higher than 8th grade in Beaufort wind scale) direction changed in a big range up to 122-degrees-angle,indicating the field measurements scoped over the typhoon landing period. 2) The ratio of the longitudinal,lateral and vertical turbulence intensities of the strong wind in the typhoon eye wall region was 1:0.96:0.36. Compared with the code defined ratio 1:0.88:0.5,the lateral component was larger and the vertical component was smaller. 3) The value of integral scale increased when the eye wall of Typhoon Nuri passed over the field measurement site. Before the center of Typhoon Nuri arrived,the integral scale of the strong typhoon wind was about twice compared with that for the non-typhoon wind. 4) The spatial correlation of the turbulent wind,coherence function curve and the decay factor had significant differences at different times during the typhoon process. In the eye wall of the typhoon,the horizontal spatial correlation was relatively strong and horizontally spatial correlation spectrum decayed slower with frequency increase. The minimum regressed coefficient C in coherence function model was 4.67,which is lower than the code defined low limit. The maximum decay factor was 27.75 which is larger than the code defined upper bound. 20Pharmacokinetics of radioimmunotherapeutic agent of direct labeling mAb ^(188)Re-HAb18显示文摘AIM: To labed Anti-hepatoma monoclonal antibody (mAb)fragment HAb18 F (ab')2 was labeled with 188 Re for thepharmacokinetic model of 188 Re-HAb18 F (ab')2 and toevaluate its pharmacokinetic parameters in hepatoma-bearing nude mice.METHODS: HAb18 F(ab')2 was directly labeled with 188Reusing 2-mercaptoethanol (2-ME) as reducing agents.Labeling efficiency and immunoreactivity of 188 Re-HAb18 F( ab')2 were evaluated by Whatman 3MM paperchromatography and live cell assay, respectively.Biodiatribution analysis was also conducted in nude micebearing human hepatoma in which animals were sacrificed atdifferent time points(1, 4, 18, 24 and 24h) after 188Re-HAb18F(ab' )2 was injected through tail-vein into hepatoma-bearingnude mice. The blood and radioactivity of organs and masswere measured. The concentrations of 188 Re-HAb18 F(ab')2were evaluated with a pharmacokinetic 3P97 software.RESULTS: The optimum labeling efficiency andimmunoreactive fraction were 91.7% and 0.78%,respectively. The parameters of 188Re-HAb18 F(ab')2 were:T1/2, 2.29h; Vd, 1.49 × 10-9L@ Bq- 1; AUC, 20.49 × 109Bq@ h@L-1 ;CL, 0.45 × 10-3L@ h- 1. 188Re- HAb18 F(ab')2 could locatespecially in hepatoma with high selective reactivity of HAb18F(ab')2. 188 Re-HAb18 F(ab')2 was mainly eliminated bykidney.
