| 1 | Twelve weeks of dance exergaming in overweight and obese adolescent girls:Transfer effects on physical activity,screen time,and self-efficacy显示文摘Background: Given the low levels of physical activity(PA) among adolescent girls in the US, there is a need to identify tools to motivate increased PA. Although there is limited evidence that adolescents transfer PA from one context to another, exergames(i.e., video games that require gross motor activity) may act as a gateway to promote overall PA outside game play. The purpose of this study was to examine potential transfer effects(i.e., influences on external behaviors and psychological constructs) of a 12-week exergaming intervention on adolescent girls' PA, screen time,and self-efficacy toward PA, as well as the intrinsic motivation of exergaming.Methods: Participants were 37 girls aged 14–18 years(65% African American, 35% white) who were overweight or obese(body mass index ≥ 85 th percentile) and were recruited from the community via school, physicians, news media, and social media websites. Adolescents were randomly assigned to a 12-week group exergaming intervention(thirty-six 60 min sessions of group-based dance exergaming in a research laboratory using Kinect for Xbox360(Microsoft Corporation, Redmond, WA, USA)) or to a no-treatment control group. Outcome variables included objectively measured PA(total) and self-reported leisure-time PA(discretionary time only) 1 week before vs. 1 week after the intervention; selected type and intensity of PA when placed in a gym setting for 30 min('cardio free choice'); screen time; self-efficacy toward PA; and intrinsic motivation toward exergaming.Results: Attendance at the exergaming sessions was high(80%). Compared with the control group, the intervention group self-reported an increase in PA(p = 0.035) and fewer hours watching television or videos(p = 0.01) after the intervention, but there were no significant differences in sedentary, light, moderate, or vigorous PA measured by accelerometry. The intervention group significantly improved self-efficacy toward PA(p = 0.028). The intervention group highly rated intrinsic motivation toward exergaming.Conclusion: Exergaming for 12 weeks was associated with positive impacts on adolescent girls' self-reported PA, television viewing, self-efficacy,and intrinsic motivation. | 2 | Reallocation of foliicolous species of the genus Strigula into six genera(lichenized Ascomycota,Dothideomycetes,Strigulaceae)显示文摘Strigula sensu lato has been previously defined based on phenotype characters as a rather broad genus including tropical to temperate species growing on a wide array of substrata.In this study,based on a multilocus phylogenetic approach,we show that foliicolous species form six well-delimited clades that correlate with diagnostic phenotype features,including thallus morphology,carbonization of the involucrellum and excipulum,ascospore dimensions,and type of macroconidia.Given the topology,with five of the six clades emerging on long stem branches,and the strong phenotypical differentiation between the clades,we recognize these at the genus level,making mostly use of previously established genus names.Four genera,namely Phylloporis,Puiggariella,Raciborskiella,and Racoplaca,are resurrected for the S.phyllogena,S.nemathora,S.janeirensis,and S.subtilissima groups,respectively,whereas one new genus,Serusiauxiella gen.nov.,is introduced for a novel lineage with peculiar macroconidia.The only sequenced non-foliicolous species,S.jamesii,is not closely related to these six foliicolous lineages but clusters with Flavobathelium and Phyllobathelium,revealing it as an additional undescribed genus-level lineage being treated elsewhere.Within the new genus Serusiauxiella,three new species are described:Serusiauxiella filifera sp.nov.,S.flagellata sp.nov.,and S.sinensis sp.nov.In addition,ten new combinations are proposed:Phylloporis austropunctata comb.nov.,P.radiata comb.nov.,P.vulgaris comb.nov.,Puiggariella confluens comb.et stat.nov.,P.nemathora comb.nov.,P.nigrocincta comb.nov.,Racoplaca maculata comb.nov.,R.melanobapha comb.nov.,R.transversoundulata,and R.tremens comb.nov.We also report on a peculiar,previously unrecognized growth behaviour of the macroconidial appendages in Strigula s.lat. | Shu‑Hua Jiang Robert Lücking Amanda Barreto Xavier‑Leite Marcela E.S.Cáceres AndréAptroot Carlos Vinas Portilla Jiang‑Chun Wei | 2020 | Fungal Diversity2020,,3: | 5 |

| 4 | Hybrid de novo genome assembly of red gromwell(Lithospermum erythrorhizon)reveals evolutionary insight into shikonin biosynthesis显示文摘Lithospermum erythrorhizon(red gromwell;zicao)is a medicinal and economically valuable plant belonging to the Boraginaceae family.Roots from L.erythrorhizon have been used for centuries based on the antiviral and woundhealing properties produced from the bioactive compound shikonin and its derivatives.More recently,shikonin,its enantiomer alkannin,and several other shikonin/alkannin derivatives have collectively emerged as valuable natural colorants and as novel drug scaffolds.Despite several transcriptomes and proteomes having been generated from L.erythrorhizon,a reference genome is still unavailable.This has limited investigations into elucidating the shikonin/alkannin pathway and understanding its evolutionary and ecological significance.In this study,we obtained a de novo genome assembly for L.erythrorhizon using a combination of Oxford Nanopore long-read and Illumina short-read sequencing technologies.The resulting genome is∼367.41 Mb long,with a contig N50 size of 314.31 kb and 27,720 predicted protein-coding genes.Using the L.erythrorhizon genome,we identified several additional phydroxybenzoate:geranyltransferase(PGT)homologs and provide insight into their evolutionary history.Phylogenetic analysis of prenyltransferases suggests that PGTs originated in a common ancestor of modern shikonin/alkanninproducing Boraginaceous species,likely from a retrotransposition-derived duplication event of an ancestral prenyltransferase gene.Furthermore,knocking down expression of LePGT1 in L.erythrorhizon hairy root lines revealed that LePGT1 is predominantly responsible for shikonin production early in culture establishment.Taken together,the reference genome reported in this study and the provided analysis on the evolutionary origin of shikonin/alkannin biosynthesis will guide elucidation of the remainder of the pathway. | Robert P.Auber Thiti Suttiyut Rachel M.McCoy Manoj Ghaste Joseph W.Crook Amanda L.Pendleton Joshua R.Widhalm Jennifer H.Wisecaver | 2020 | Horticulture Research2020,7,1: | 2 |
| 5 | More forks on the road to replication stress recovery显示文摘High-fidelity replication of DNA,and its accurate segregation to daughter cells,is critical for maintaining genome stability and suppressing cancer.DNA replication forks are stalled by many DNA lesions,activating checkpoint proteins that stabilize stalled forks.Stalled forks may eventually collapse,producing a broken DNA end.Fork restart is typically mediated by proteins initially identified by their roles in homologous recombination repair of DNA double-strand breaks(DSBs).In recent years,several proteins involved in DSB repair by non-homologous end joining(NHEJ)have been implicated in the replication stress response,including DNA-PKcs,Ku,DNA Ligase IV-XRCC4,Artemis,XLF and Metnase.It is currently unclear whether NHEJ proteins are involved in the replication stress response through indirect(signaling)roles,and/or direct roles involving DNA end joining.Additional complexity in the replication stress response centers around RPA,which undergoes significant post-translational modification after stress,and RAD52,a conserved HR protein whose role in DSB repair may have shifted to another protein in higher eukaryotes,such as BRCA2,but retained its role in fork restart.Most cancer therapeutic strategies create DNA replication stress.Thus,it is imperative to gain a better under-standing of replication stress response proteins and pathways to improve cancer therapy. | Chris Allen Amanda K.Ashley Robert Hromas Jac A.Nickoloff | 2011 | Journal of Molecular Cell Biology2011,3,1: | 2 |
