1Metabonomic phenotype and identification of “heart blood stasis obstruction pattern” and “qi and yin deficiency pattern” of myocardial ischemia rat models显示文摘The traditional Chinese medicine concepts of 'Xinxueyuzuzheng (heart blood stasis obstruction pattern)' and 'Qiyinliangxuzheng (qi and yin deficiency pattern)' for myocardial ischemia rat models were constructed in the present study. Endogenous metabolites in rat plasma were analyzed using the GC/TOF-MS-based metabonomic method. Significant metabolic differences were observed between the control and two model groups, and the three groups were distinguished clearly by pattern recognition. Compared with those of the control, the levels of hydroxyproline, threonic acid, glutamine and citric acid were strikingly up or down-regulated in model rats. The metabolites contributing most to the classification between the two 'pattern' rats were identified, such as valine, serine, threonine, ornithine, hydroxyproline, lysine, 2-hydroxybutanoic acid, 3-hydroxybutanoic acid, galactofuranose and inositol. These compounds were indicated as the potential biomarkers. The results suggested that the two 'patterns' are involved in dysfunction in oxidative stress, energy metabolism and amino acid metabolism. 2Effects of hypoxia,hyperoxia on the regulation of expression and activity of matrix metalloproteinase-2 in hepatic stellate cells显示文摘AIM To study the effects of hypoxia, hyperoxia on the regulation of expression and activity of matrix metalloproteinase-2 (MMP-2) in hepatic stellate cells ( HSC).``METHODS The expressions of MMP-2, tissue inhibitor of mjatrix metalloproteinese-2 ( TIMP-2 ) and membrane type matrix metalloproteiness-l (MT1-MMP) in cultured rat HSC were detected by immunocytochemistry (ICC) and in situ hybridization (ISH). 3Diagnosis and treatment of congenital choledochal cyst: 20 years' experience in China显示文摘AIM To summarize the experience of diagnosis and treatment of congenital choledochal cyst in the past 20years ( 1980 2000).``METHODS The clinical data of 108 patients admitted from 1980 to 2000 were analyzed retrospectively.RESULTS Abdominal pain, jaundice and abdominal mass were presented in most child cases. Clinical symptoms in adult cases were non-specific, resulting in delayed diagnosis frequently. Fifty-seven patients (52.7%) had coexistent pancreatiobiliary disease. Carcinoma of the biliary duct occurred in 18 patients (16.6%). Ultrasonic examination was undertaken in 94 cases, ERCP performed in 46 cases and CT in 71 cases. All of the cases were correctly diagnosed before operation. Abnormal pancreatobiliary duct junction was found in .39 patients.Before 1985 the diagnosis and classification of congenital choledochal cyst were established by ultrasonography preoperatively and confirmed during operation, the main procedures were internal drainage by cyst enterostomy.After 1985, the diagnosis was established by ERCP and CT. and cystectomy with Roux-en-Y hepaticojejunostomy was the conventional procedures. In 1994, we reported a new and simplified operative procedure in order to reduce the risk of choledochal cyst malignancy. Postoperative complication was mainly retrograde infection of biliary tract, which could be controlled by the administration of antibiotics, there was no perioperative mortality.``CONCLUSION The concept in diagnosis and treatment of congenital choledochal cyst has obviously been changed greatly. CT and ERCP were of great help in the classification of the disease. 4Deep seismic reflection profiling revealing the complex crustal structure of the Tongling ore district显示文摘The deep seismic profiling across the Tongling ore district reveals a complex crustal structure. Strong contrasting dipping layered reflections (4—11 s, TWT),which dominate the lower crust of the northern part of the profile, are interpreted as the underplating of the basalt related to extensional tectonism. In the south of the profile, the Yangtze craton is characterized by strong reflections in the middle crust, showing a distinct two-layer crustal structure. Over the region of the Tongling uplift, there appear the complex arc shape reflections, suggesting folded, faulted and intruded structures, and the transparent zone below them reveals the existence of batholith. The south dipping strong reflections between the upper crust and lower crust (4—7 s, TWT) suggest a detachment between them. The detachment provided space for the magma intrusion, and caused the formation of the batholith. The Yangtze craton has a clear Moho re-flection, while the Tongling uplift has a weak Moho, whereas below the reflective lower crust in the northern part of the profile, there are sub-Moho reflections. 5Abnormal expression of HSP gp96 associated with HBV replication in human hepatocellular carcinoma显示文摘BACKGROUND: Heat shock protein (HSP) gp96 is a member of the HSP90 family and presumably overexpresses as a result of stimulation by mutated or abnormal proteins. Its abnormal expression correlates with carcinogenesis, progression and prognosis of hepatocellular carcinoma (HCC). In this study, we investigated the pathological characteristics of liver gp96 expression and its relationship with hepatitis B virus (HBV) replication in HCC patients. METHODS: Tumor specimens were prospectively collected from 30 HCC patients undergoing liver resection. Total RNAs were extracted from HCC or their noncancerous tissues. The distribution of gp96 expression in hepatocytes was investigated by streptavidin peroxidase (S-P) immunohistochemistry and tissue HBV-DNA was detected by the in situ molecular hybridization technique. The association of gp96 expression with HBV replication, and the histopathological characteristics of HCC were analyzed. RESULTS: The gp96 was strongly expressed in HCC (73.3%, 22 of 30) and weakly (46.7%, 14 of 30) in non-cancerous tissues. The gp96 expression in HCC tissues was correlated with degree of tumor differentiation and tumor size, but not with tumor number (P>0.05). Immunohistochemical analysis showed that 17 of 19 HCC patients with HBVDNA -positive were strongly expressed for gp96, whereas only 5 of 11 patients with HBV-DNA-negative were positive for gp96. A significant difference was found between the two groups (89.5% vs. 45.5%, P<0.05). CONCLUSIONS: The abnormal expressions of HSP gp96 in HCC tissues are associated with HBV replication. 7Sand-dust storms in China: temporal-spatial distribution and tracks of source lands显示文摘Sand-dust storm is a special natural disaster that frequently occurs in deserts and their surrounding areas. With the data published on Surface Meteorological Monthly Bulletin and Surface Chart during 1971-1996, the temporal-spatial distribution and annual variation of sand-dust storms are analyzed on the basis of the case study of atmospheric processes. Furthermore, the tracks and source areas of sand-dust storms are determined with the aid of GIS. The results show that except some parts of Qinghai Province and Inner Mongolia as well as Beijing, sand-dust storms decrease apparently in time and space in recent decades in China. Sand-dust storms occur most frequently in spring, especially in April. According to their source areas, sand-dust storms are classified into two types, i.e., the inner-source and outer-source sand-dust storms. Most of the outer-source sand-dust storms move along the north and west tracks. The north-track outer-source sand-dust storms always intrude into China across the Sino-Mongolian border from Hami, a city in the eastern part of Xinjiang, to Xilin Gol, a league in Inner Mongolia, while the west-track ones intrude into China from both southern and northern Xinjiang. 8Recombinant expression of rt-PA gene (encoding Reteplase) in gametophytes of the seaweed Laminaria japonica (Laminariales,Phaeophyta)显示文摘The life cycle of seaweed Laminaria japonica involves a generation alternation between diploid sporophyte and haploid gametophte. The expression of foreign genes in sporophte has been proved. In this research, the recombinant expression in gametophyte was investigated by particle bombardment with the rt-PA gene encoding the recombinant human tissue-type plasminogen activator (Reteplase), which is a thrombolytic agent for acute myocardial infarction (AMI). Transgenic gametophytes were selected by their resistance to herbicide phosphiothricin (PPT), and proliferated in an established bubble column photo-bioreactor. 10The Mitochondrial DNA Mutation at Position 11778 in Chinese Families with Leber's Hereditary Optic Neuropathy显示文摘We amplified the 340 bp of mitochondrial DMA (mtDNA) by PCR including the recognized sequence of restriction enzyme of SfaN I . After amplification and digestion of SfaN I , two bands of 190 bp and 150 bp appeared in the mtDNA of four normal individuals but only one band of 340 bp appeared in the mtDNA with the mutation of G to A at the site of the nucleotide 11778 because such mutation destroyed the recognized sequence of SfaN I . 12Preparation and characterization of polyclonal antibodies against ARL-1 protein显示文摘AIM: To prepare and characterize polyclonal antibodies against aldose reductase-like (ARL-1) protein.METHODS: ARL-L gene was inserted into the E.coli expression vector pGEX-4T-1(His)6C and vector pQE-30. Recombinant ARL1 proteins named ARL-(His)6 and ARL-GST were expressed.They were purified by affinity chromatography. Sera from domestic rabbits immunized with ARL-(His)6 were purified by CNBr-activated sepharose 4B coupled ARL-GST. Polyclonal antibodies were detected by Western blotting.RESULTS: Recombinant proteins of ARL-(His)6 with molecular weight of 35.7 KD and ARL-GST with molecular weight of 60.8 KD were highly expressed. The expression levels of ARL-GST and ARL-(His)6 were 15.1% and 27.7 %among total bacteria proteins, respectively. They were soluble, predominantly in supernatant. After purification by non-denatured way, SDS-PAGE showed one band. In the course of polyclonal antibodies purification, only one elution peak could be seen. Western blotting showed positive signals in the two purified proteins and the bacteria transformed with pGEX-4T-1(His)6 C-ARL and pQE-30-ARL individually.CONCLUSION: Polyclonal antibodies are purified and highly specific against ARL-1 protein. 13Adverse effects of metal exposure on chemotaxis towards water-soluble attractants regulated mainly by ASE sensory neuron in nematode Caenorhabditis elegans显示文摘Chemotaxis to water-soluble attractants is mainly controlled by ASE sensory neuron whose specification is regulated by che-1 in Caenorhabditis elegans. Our data suggested that exposure to high concentrations of metals, such as Pb, Cu, Ag, and Cr, would result in severe defects of chemotaxis to water-soluble attractants of NaCl, cAMP, and biotin. Moreover, the morphology of ASE neuron structures as observed by relative fluorescent intensities and relative size of fluorescent puncta of cell bodies, relative lengths of sensory endings in ASE neurons, and the expression patterns of che-1 were obviously altered in metal exposed animals when they meanwhile exhibited obvious chemotaxis defects to water-soluble attractants. In addition, the dendrite morphology could be noticeably changed in animals exposed to 150 μmol/L of Pb, Cu, and Ag. Furthermore, we observed significant decreases of chemotaxis to water-soluble attractants in Pb exposed che-1 mutant at concentrations more than 2.5 μmol/L, and in Cu, Ag, and Cr exposed che-1 mutant at concentrations more than 50 μmol/L. 15Pharmacokinetic Study of Baicalein and Its Major Metabolites after iv Administration in Dogs显示文摘Objective To develop and validate a simple,rapid,sensitive,and reproducible HPLC method for simultaneous determination of baicalein and its metabolite baicalin in dog plasma and for the subsequent pharmacokinetic study after iv administration to dogs.Methods An accurate and reproducible HPLC-UV method was developed and validated for simultaneous determination of baicalein and baicalin in dog plasma,using luteolin as internal standard.The analytes were separated by an Agilent Zorbax SB-C18 column(250 mm × 4.6 mm,5 μm) and the column temperature was maintained at 40 ℃.The mobile phase was a binary mixture of acetonitrile and water(27:73) ,containing 0.05% phosphoric acid in water,with a flow rate of 1.0 mL/min.The UV detector was set at 276 nm.Results Linear relationships were validated over the range of 0.05-25 μg/mL for baicalein and 0.05-20 μg/mL for baicalin.The intra-and inter-day precision values for all samples were within 8.0%,using relative standard deviation.This method was successfully applied to the pharmacokinetic studies in dogs after iv administration of baicalein.Baicalein was converted to baicalin quickly.Cmax values were 21.13 μg/mL at 0.05 h for baicalein and 1.57 μg/mL at 0.5 h for baicalin,areas under the plasma concentration-time curve were 4.97 h·μg/mL for baicalein and 0.63 h·μg/mL for baicalin,and the elimination half-life is 0.50 h for baicalein and 0.75 h for baicalin,respectively.Conclusion The method is able and sufficient to be used in drug metabolism and pharmacokinetic studies of baicalein.TIAN Shuo1,DU Li-da2,WANG Shou-bao1,HE Guo-rong1,YANG Tao1,LI Xiao-xiu1,GUO Jing1,DU Guan-hua1 1.National Centre for Pharmaceutical Screening,Institute of Materia Medica,Chinese Academy of Medical Sciences and Peking Union Medical College,Beijing 100050,China 2.School of Pharmacy,China Pharmaceutical University,Nanjing 210009,China 2011Chinese Herbal Medicines2011,3,3:5
16Hydroisomerization of n-heptane over bimetal-bearing H_3PW_(12)O_(40) catalysts supported on dealuminated USY zeolite显示文摘The bimetal-bearing (CePt or LaPt) 12-tungstophosphoric acid (H3PW12O40 (PW)) catalysts supported on dealuminated USY zeolite (DUSY) were prepared by impregnation and characterized by XRD, BET, IR, and H2-chemisorption. Their catalytic activities were tested in the hydroisomerization of n-heptane with a continuous atmospheric fixed-bed reactor. After the steam treatment combined with the acid leaching, as well as the supporting with PW and the bimetals, the DUSY support retains the Y zeolite porosity and the PW well keeps its Keggin structure in catalysts. The doping of Ce into the catalysts enhances the dispersion of Pt on the catalyst surface. The Pt-bearing PW catalysts doped with Ce or La, especially Ce, exhibit much higher catalytic activity and selectivity than the catalysts without dopants at lowered reaction temperatures. 18Pyridine-H_5PMo_(10)V_2O_(40) hybrid catalysts for liquid-phase hydroxylation of benzene to phenol with molecular oxygen显示文摘Pyridine(Py)-modified Keggin-type vanadium-substituted heteropoly acids(PynPMo10V2O40,n=1 to 5) were prepared by a precipitation method as organic/inorganic hybrid catalysts for direct hydroxylation of benzene to phenol in a pressured batch reactor and their structures were detected by FT-IR.Among various catalysts,Py3PMo10V2O40 exhibits the highest catalytic activity(yield of phenol,11.5%),without observing the formation of catechol,hydroquinone and benzoquinone in the reaction with 80 vol% aqueous acetic acid,molecular oxygen and ascorbic acid used as the solvent,oxidant and reducing reagent,respectively.Influences of reaction temperature,reaction time,oxygen pressure,amount of ascorbic acid and catalyst on yield of phenol were investigated to obtain the optimal reaction conditions for phenol formation.Pyridine can greatly promote the catalytic activity of the Py-free catalyst(H5PMo10V2O40),mostly because the organic π electrons in the hybrid catalyst may extend their conjugation to the inorganic framework of heteropoly acid and dramatically modify the redox properties,at the same time,pyridine adsorbed on heteropoly acids can promote the effect of 'pseudo-liquid phase',thus accounting for the enhancement of phenol yield.GE HanQing1,LENG Yan1,ZHANG FuMin1,PIAO JiaRui2,ZHOU ChangJiang1 & WANG Jun1 1 State Key Laboratory of Materials-Oriented Chemical Engineering,College of Chemistry and Chemical Engineering,Nanjing University of Technology,Nanjing 210009,China 2 Petrochemical Research Institute,PetroChina Company Ltd.,Beijing 100083,China 2009Science China Chemistry2009,52,8:4

20A New Enolate Furostanoside from Asparagus Filicinus显示文摘A new enolate derivative of furostanol glycoside, named asparagusin A, was isolated from the roots of Asparagus filicinus and established as 3 - O- 6 - D- xylopyranosyl (l →4)- β - Dglucopyranosyl (25S)-furost-20(22)-ene-3β, 26-diol 26-O-0-D-glucopyranoside (l) by spectroscopic and chemical methods.
