| 1 | Effect of bax,bcl-2 and bcl-xL on regulating apoptosis in tissues of normal liver and hepatocellular carcinoma显示文摘AIM: To investigate the expression of bax, bcl-2 and bcl-xL mRNA in the tissues of normal liver and hepatocellular carcinoma (HCC), and analyze the relationship between the expression of bax, bcl-2 and bcl-xL mRNA and clinical parameters of HCC patients.METHODS: The expression of bax, bcl-2 and bcl-xL mRNA of normal liver and HCC was measured by Northern blot. Statistical analyses were made by t test and correlation analysis.RESULTS: A very low mRNA level was indicated at bax,bcl-2 and bcl-xL in the HCC tissues in contrast to the tissues of normal liver by Northern blot analysis. The analyses of mRNA level revealed that HCC tissues exhibited a mean 7.6-fold decrease in bax, 4.2-fold in bcl-2 and 3.5-fold in bcl-xL in comparison with normal control tissues, respectively. Positive correlation was found between bax and bcl-xL (r=0.7061,P<0.01). There was no significance between the mRNA expression of these three genes and age, gender, tumor differentiation and tumor stage of HCC patients. | 2 | Effect of Cultivation on Spatial Distribution of Nematode Trophic Groups in Black Soil显示文摘Geostatistics combined with GIS was applied to assess the spatial distribution of nematode trophic groups following two contrasting soil uses in the black soil region of Northeast China. Two plots, one with fallow for 12 years and the other cultivated, were marked on regular square grids with 2-m spacing. Soil samples were collected from each sampling point, nematodes were extracted from these samples and classified into four trophic groups: bacterivores, fungivores, plant parasites, and omnivores/predators. The numbers of total nematodes and trophic groups analyzed had normal distributions on both fallow and cultivated plots. The absolute abundances of total nematodes and trophic groups were observed to be much more homogeneous on cultivated plot than on fallow one. Geostatistical analysis showed that the densities of total nematodes and trophic groups on both fallow and cultivated plots exhibited spatial dependence at the sampled scale and their experimental semivariograms were adjusted to a spherical or exponential model, except those of bacterivores and fungivores on cultivated plot. | 3 | Fractionation of heavy metals in shallow marine sediments from Jinzhou Bay,China显示文摘This work investigated the distribution and speciation of Cd, Cu, Pb, Fe and Mn in the shallow sediments of Jinzhou Bay, Northeast China, which has been heavily contaminated by nonferrous smelting activities. The concentrations of Cd, Cu and Pb in sediments were found to be 100, 13 and 7 times, respectively, being higher than the national guideline (GB 18668-2002). Sequential extraction test showed that 39%-61% of Cd were exchangeable fractions, indicating that Cd in the sediments posed a high risk to local environments. While Cu and Pb were at moderate risk levels. | 5 | Rapid ecotoxicological assessment of heavy metal combined polluted soil using canonical analysis显示文摘Quick, simple to perform, and cheap biomarkers were combined in a rapid assessment approach to measure the effects of metal pollutants, Cu, Cd, Pb and Zn in meadow burozem on wheat. Analysis of orthogonal design showed that the significant zinc factor indicated both the inhibition rate of shoot mass and that of root elongation were affected by zinc(P<0.05 and P<0.01, respectively). The first toxicity canonical variable (TOXI), formed from the toxicity data set, explained 49% of the total variance in the toxicity data set; the first biological canonical variable(BIOL) explained 42% of the total variation in the biological data set. The correlation between the first canonical variables TOXI and BIOL (canonical correlation) was 0.94 (P<0.0001). Therefore, it is reliable and feasible to use the achievement to assess toxicity of heavy metal combined polluted soil using canonical analysis. Toxicity of soil combined polluted by heavy metals to plant community was estimated by comparing the IC 50 values describing the concentration needed to cause 50% decrease with grow rate compared to no metal addition. Environmental quality standard for soils prescribe that all these tested concentration of heavy metals in soil should not cause hazard and pollution ultimately, whereas it indicated that the soils in second grade cause more or less than 50% inhibition rates of wheat growth. | 7 | Meteorological control on CO_2 flux above broad-leaved Korean pine mixed forest in Changbai Mountains显示文摘The impacts of temperature, photosynthetic active radiation (PAR) and vapor pressure deficit (VPD) on CO2 flux above broad-leaved Korean pine mixed forest in the Changbai Mountains were studied based on eddy covariance and meteorological factors measurements.The results showed that, daytime CO2 flux was mainly controlled by PAR and they fit Michaelis-Menten equation. Meanwhile VPD also had an influence on the daytime flux. Drier air reduced the CO2 assimilation of the ecosystem, the drier the air, the more the reduction of the assimilation. And the forest was more sensitive to VPD in June than that in July and August. The respiration of the ecosystem was mainly controlled by soil temperature and they fit exponential equation. It was found that this relationship was also correlated with seasons; respiration from April to July was higher than that from August to November under the same temperature. Daily net carbon exchange of the ecosystem and the daily mean air temperature fit exponential equation. It was also found that seasonal trend of net carbon exchange was the result of comprehensive impacts of temperature and PAR and so on. These resulted in the biggest CO2 uptake in June and those in July and August were next. | 8 | Expression of p57^(kip2), Rb protein and PCNA and their relationships with clinicopathology in human pancreatic cancer显示文摘AIM: To investigate the effects of inhibiting factor of cellcycle regulation p57kip2, retinoblastinoma protein (Rb protein)and proliferating cell nuclear antigen (PCNA) in the genesisand progression of human pancreatic cancer.METHODS: The expression of p57kip2, Rb protein and PCNAin tumor tissues and adjacent tissues of 32 patients withpancreatic cancer was detected with SP immunohistochemicaltechnique.RESULTS: p57kip2 protein positive-expression rate in tumortissues of pancreatic cancer was 46.9 %, which was lowerthan that in adjacent pancreatic tissues (75.0 %) (x2=5.317,P<0.05), p57kip2 protein positive-expression correlatedsignificantly with tumor cell differentiation (well-differentiationversus moderate or low-differentiation, P<0.05) but did notcorrelate significantly with lymph node metastasis (lymph nodemetastasis versus non-lymph node metastasis, P>0.05); Rbgene protein positive-expression rate in tumor tissues was50.0 %, which was also lower than that in adjacent pancreatictissues (78.1%) (x2=5.497, P<0.05); PCNA positive-expression rate was 71.9 %, being higher than that inadjacent pancreatic tissues (43.8 %) (x2=5.189, P<0.05),PCNA positive-expression also correlated significantly withtumor cell differentiation and lymph node metastasis (well-differentiation versus moderate or low- differentiation, lymphnode metastasis versus non-lymph node metastasis, P<0.05).Rb protein positive-expression rate in the tumor tissues ofp57kip2 protein positive-expression group was 53.3 %; andRb protein positive-expression rate in the tumor tissues ofp57kip2 protein negative-expression group was 47.1%. | 9 | Induction of apoptosis by furanodiene in HL60 leukemia cells through activation of TNFR1 signaling pathway显示文摘Objective Curcuma wenyujin,named Ezhu in Chinese,a traditional Chinese medicine,which has been shown to possess anticarcinogenic activity and used for the treatment of tumor in China,for example,hepatic cancer and leukemia.β-elemene,a component of Curcuma wenyujin,was largely reported as a main active component for anti-tumor effect of Curcuma wenyujin.However,furanodiene,another main component of Curcuma wenyujin,was seldom investigated for its anti-tumor activities.In the present study,we aim to investigate the effect of furanodiene on human leukemia HL60 cells and to study the accurate molecular mechanisms of its action.Methods Trypan blue exclusion experiment was used for cell growth inhibition assay.The apoptotic characterizations were assessed by flow cytometry analysis,AO/EB staining assay and agarose gel electrophoresis assay.The expression of apoptosis-related proteins and the release of cytochrome c from mitochondrial were detected by western blotting,and the mRNA levels of TNF-α and TNFR1 were probed by RT-PCR.Formation of TNFR1 complex was analyzed by using immunoprecipitation of TNFR1 with TRRF1/2 and RIP.Results Furanodiene(10-100 μM)treatment inhibited the growth of HL60 cells in a concentration-dependent manner.The effect of furanodiene on HL60 cells was associated with the induction of apoptosis,which was characterized by DNA fragmentation,cleavage of poly(ADP-ribose)polymerase(PARP),caspase-3,caspase-8 and caspase-9.In the Bcl-2 family proteins,Bid protein(a substrate of caspase-8)was activated by furanodiene,but Bcl-2,Bax and Bcl-xL proteins were not inuenced by furanodiene stimulation.Furanodiene elicited cytochrome c release from mitochondria into the cytosol.Moreover,furanodiene treatment caused the upregulation of TNFR1,the formation of TNFR1 complex and an obvious production of TNF-α in HL60 cells.The soluble TNFR1 receptor effectively inhibited furanodiene-induced apoptosis.Conclusions Furanodiene inhibited the growth of HL60 leukemia cells via induction of apoptosis.Furanodiene-induced apoptosis in HL60 cells is mediated by upregulation of TNF receptor 1 as well as induction of TNF-α production to activate TNFR1 signaling pathway.Our research provides insight into the molecular mechanisms on furanodiene-induced cell death,and may aid to the development of furanodiene as a new anti-tumor agent. | MA En-long1,WANG Xiao-long1,LI Yan-chun1,TAI Wen-jiao1,LI Te1,GUO Tao2(1.Department of Pharmacology,Shenyang Pharmaceutical University,Shenyang 110016,China 2.Department of Pharmacy,General Hospital of Shenyang Military Region,Shenyang 110016,China) | 2008 | 沈阳药科大学学报2008,25,S1: | 14 |

Under doubled CO2 concentration, grain yield and biomass increased, the SOD activity, vitamin C (Vc) and carotenoid (Car) content also increased; Superoxide (O2-.) generating rate decreased, relative conductivity and malondialdehyde (MDA) content significantly declined.But under doubled O3 concentration, the SOD activity, Vc and Car contents declined, resulting in imbalance of activated-oxygen production, enhanced O2-. generating rate and accelerated process of lipid peroxidation and increase in MDA content and ion leakage of leaves. The final result was decreased grain yield and plant biomass. Interactive effects of doubled CO2 and O3 concentrations on soybean were mostly counteractive. | 11 | Spatial heterogeneity of DTPA-extractable zinc in cultivated soils induced by city pollution and land use显示文摘The spatial heterogeneity of DTPA-extractable zinc in the cultivated soils of Shenyang suburbs in Liaoning Province of China was investigated, and its map was drawn by the methods of geostatistics combined with geographic information system. The data of soil DTPA-extractable zinc fitted normal distribution after logarithm transformation, and its semivariogram fitted a spherical model. The semivariogram indicated that the spatial dependence of soil DTPA-extractable zinc content was moderate, with the spatial dependence range of 1.69 km and the fractal dimension of 1.96. Stochastic factors contributed to 49.9% of the spatial variability, while structural factors contributed to 50.1% of it. The spatial heterogeneity of soil DTPA-extractable zinc shown by a kriged interpolation map was deeply influenced by stochastic factors such as city pollution, land use pattern and crop distributions. For example, the average content of Zn in vegetable garden soils was 2.5-4 times as much as in their originated soils, and was lower in paddy soils than in their originated soils. The areas with a higher content of soil DTPA-extractable zinc appeared in the near suburbs and the riverside along Hunhe River and the wastewater drainage of Xihe River, and the extremely high values in the near suburb of the city's residential area were a striking feature, indicating the key role of city pollution in the spatial heterogeneity of soil DTPA-extractable zinc. | 13 | Seasonal patterns and environmental control of ecosystem respiration in subtropical and temperate forests in China显示文摘Continuous measurement of carbon dioxide exchange using the eddy covariance (EC) technique was made at two ChinaFLUX forest sites including the young subtropical Pinus plantation (Qianyanzhou) and old temperate broad-leaved Korean pine mixed forest (Changbai Mountains) as part of the ChinaFLUX network. Seasonal patterns and environmental control of ecosystem respiration in the subtropical and temperate forests were evaluated by the often-used multiplicative model and Q10 model as a function of temperature and soil water content. The resuits suggested that ( i ) temperature was found to be a dominant factor in the ecosystem respiration, and most of the temporal variability of ecosystem respiration was explained by temperature. However, in the drought-stressed ecosystem, soil water content controlled the temporal variability of ecosystem respiration other than temperature effects, and soil water content became a dominat factor when severe drought affected the ecosystem respiration; (ii) the regression models analysis revealed that in the drier soil, ecosystem respiration was more sensitive to soil moisture than was expressed by the often-used multiplicative model. | 14 | 316L Austenite Stainless Steels Strengthened by Means of Nano-scale Twins显示文摘By means of dynamic plastic deformation (DPD) followed by thermal annealing, a mixed structure of micro-sized austenite grains embedded with nano-scale twin bundles (of about 20% in volume) has been synthesized in a 316L stainless steel (SS). Such a 316L SS sample exhibits a tensile strength as high as 1001 MPa and an elongation-to-failure of about 23%. The much elevated strength originates from the presence of a considerable number of strengthening nano-twin bundles, while the ductility from the recrystallized grains. The superior strength-ductility combination achieved in the nano-twins-strengthened austenite steel demonstrates a novel approach for optimizing the mechanical properties in engineering materials. | G.Z. | 15 | Electrochemical Corrosion Behavior of Nanocrystalline Materials-a Review显示文摘Nanocrystallization significantly infiuences the electrochemical corrosion behaviors of metals/alloys in liquid system. In active dissolution, nanocrystallization accelerates the corrosion reactions. If the corrosion products are dissoluble, the corrosion rate is increased by nanocrystallization; if the corrosion products are insoluble, the corrosion rate is decreased on the contrary because the corrosion products act as a block layer to delay the dissolution. In passivation, nanocrystallization changes the composition of the passive film, and results into different morphology and growth process of the passive film, both of which improves the formation of compact film and influences the semiconductor property. It influences the passivation depending on fast element diffusion and special adsorbed ability. The small grain size improves the element diffusion, which leads to the different composition of passive film (passive elements enrichment such as Cr, Ti). The small grain size also changes the surface condition, which influences the ions adsorption. All increase the corrosion resistance of materials. In local corrosion, nanocrystallization increases the unstable points on the surface of the materials, which increases the possibility of local corrosion. | 16 | Artificial coordinating field and its application to motion planning of robots in uncertain dynamic environments显示文摘Artificial coordinating fields (ACF) are proposed to deal with the motion planning problems of mobile robots in uncertain dynamic environments. An ACF around an obstacle can generate two orthogonal force vectors to a robot: one is called the coordinating force vector which is purposively designed in this paper, and the other is the repulsive force vector which is the same as that in a conventional artificial potential field. The ACF is designed according to the updated motion purpose and the relative states of the robot with respect to its local environment, and it also satisfies the robots dynamic constraints. The direction of the coordinating force can be determined on line according to an optimal evaluation function. The ACF can effectively remove the local minima, and reduce the oscillation of the planned trajectory between multiple obstacles. Only local knowledge of the environments is needed in the ACF-based motion planning. The properties of the ACF such as controllability, adaptability, safety and reachability are studied and discussed in detail in this paper. | 17 | Nickel-free Stainless Steel for Medical Applications显示文摘BIOSS4 steel is essentially a nickel-free austenitic stainless steel developed by the Institute of Metal Research, Chinese Academy of Sciences, in response to nickel allergy problems associated with nickel-containing stainless steels that are widely used in medical applications. The high nitrogen content of this steel effectively maintains the austenitic stability and also contributes to the high levels of corrosion resistance and strength. BIOSS4 steel possesses a good combination of high strength and toughness, better corrosion resistance, and better blood compatibility, in comparison with the medical 316L stainless steel. | 18 | KAI1 inhibits HGF-induced invasion of pancreatic cancer by sphingosine kinase activity显示文摘BACKGROUND:KAI1/CD82 has been reported to attenuate the process of metastases in a variety of tumors;however,its mechanism of action in invasion has not been fully elucidated. The present study aimed to investigate the importance of KAI1 in invasion and its correlation with activation of sphingosine kinase(SPK)in human pancreatic cancer PANC1 and Miapaca-2 cell lines. METHODS:The expression of KAI1 in PANC1 and Miapaca-2 cells,which was mediated by recombinant adenovirus(Ad-KAI1), was assessed by a flow cytometer and Western blotting.After successful infection was established,in vitro growth curve and invasive ability in Boyden Chamber assay were studied.The presence of KAI1 correlating with c-Met and SPK was detected by co-immunoprecipitation and[γ-32P]ATP incorporation. RESULTS:KAI1 genes had no significant effects on the curve representing cell growth.After infection with the KAI1 gene,decreased invasive ability in the Boyden Chamber assay was observed in PANC1 and Miapaca-2 cells that were induced by hepatocyte growth factor.Over-expression of KAI1 in the cells led to the deactivation of SPK and a decreased level of intracellular sphingosine-1-phosphate.No correlation was observed between c-Met and KAI1 during co-immunoprecipitation. Because of the complexity of forest ecosystems, the applications of FEM can be facilitated with decision support systems (DSS) that recognize and incorporate ecological and socio-economic variables. With the rapid development of computation and information technologies, DSS have been advanced in many ways. Traditional forest management within a forestry unit in China is planned on a yearly basis. The planning itself remains primarily a verbal concept as there are no quantitative decision-support tools available to translate the concept into forest management actions. For the purposes of FEM at the management level, a forest management DFF, FORESTAR(R), has been developed under a framework of geographic information system (GIS) and forest models. | 20 | Changes of Oxygen Content in Molten TiAl Alloys as a Function of Superheat during Vacuum Induction Melting显示文摘Experimental studies on the melting process of titanium aluminum alloy have been pursued from the viewpoint of contamination.TiAl alloys were prepared with vacuum induction melting (VIM) in calcia crucibles at 1873 K and 1923 K in order to determine the behavior of the oxygen content as a function of temperature,time and frequency of power.The experiment results showed that alloys were uncontaminated except for the increasing of oxygen content which was introduced from the reaction CaO(s)=Ca(in TiAl)+O(in TiAl),and the standard Gibbs energy of the reaction was determined to be △G0 =274000 102.8T(J/mol).Oxygen content increased slowly with the melting time by about 50×10-6 wt pct/min,and decreased with induction melting frequency.Lower superheat and higher melting frequency can be used to reduce oxygen content increasing rate. | Bo Chen,Yingche Ma,Ming Gao and Kui Liu Institute of Metal Research,Chinese Academy of sciences,Shenyang 110016,China | 2010 | Journal of Materials Science & Technology2010,26,10: | 8 |
